Works as expected
Review Date: 1 Aug 2024 | Biostatus Limited
Very simple use to detect the nuclei and the signal is generally bright.
To visualize nuclei for basic research
It is easy to use and the staining is generally bright. In my case it was helpful since it was not possible to detect the fluorescence released by other compounds used to visualize nuclei such as dapi or hoechst 33342.

A bacterial DNA DRAQ5 was added to differentiate them from the background.
labeling bacterial viability for flow cytometry and confocal microcopy analysis
We ordered the product from Thermo Fisher. It is very easy to use in an effective and economical concentration range (after titration). Its use on fixed bacteria is a success. The protocol we have developed is the subject of a technical note: A flow cytometry method for safe detection of bacterial viability (DOI: 10.1002/cyto.a.24794)

Biostatus: Single cell transcriptomics and genomics
In this whitepaper, Biostatus Limited examines sample preparation and the concerns over quality assurance of cell delivery for downstream analysis. This whitepaper highlights that sample preparation will improve quality and validity of the molecular analysis when performed to a high standard.
Using DRAQ5™ in HCS – What Can You See?
DRAQ5™ has been used widely in high content screening and related drug discovery assays for more than a decade. Recent papers give a clear overview of the different intracellular features one can render with DRAQ5™ staining.
Application of DRAQ™ Dyes in Single Cell Transcriptomics and Genomics
DRAQ5™ and DRAQ7™ dyes provide accurate fluorescent labelling and assist in distinguishing cell status for single cell transcriptomics and genomic applications. One concern regarding the use of these DNA binding dyes is any unwanted effects on the first strand cDNA synthesis. However, studies in this white paper have demonstrated that DRAQ5™ and DRAQ7™ do not interfere with the synthesis process, indicating an accurate experimental outcome.
DRAQ5™ in FC-Based Apoptosis Assays: Far-Red Fluorescent Live-Cell Permeant DNA Dye
The cell cycle is a series of events that occur when a cell divides and replicates into two daughter cells. Cell cycle regulation depends upon processes that are pivotal to cell survival. During the cell cycle the quantity of DNA increases from 2N (G1 phase) to 4N (G2 phase) via the S (synthesis) phase. Perturbation of the cell cycle can lead, for example, to cell cycle arrest or uncontrolled cell division, as in cancer, while cells with DNA below 2N are typically in apoptosis, this is the, so-called, sub-G1/G0 peak. The quantity of DNA in each nucleated cell in a population can be measured by labeling the nuclear DNA with a fluorescing DNA dye, most commonly propidium iodide (PI). As a live-cell permeant DNA probe DRAQ5™ can be applied directly to complex samples such as blood or bone marrow or cultured cells, as the last step prior to analysis and without any washing steps.
APOPTRAK™ in FC-Based Apoptosis Assays: Live Cell Marking and Cell Death Scoring
In apoptosis an ordered series of events leads to destruction of a single cell. The changes that occur can be assayed through DNA fragmentation, caspase activity (using permeant substrates e.g. FLICA™ probes), mitochondrial membrane potential (e.g. using TMRM), cytochrome C release, plasma membrane inversion (using Annexin V) and permeabilization being common choices. This application note demonstrates the use of APOPTRAK™ in the analysis of far-red fluorescent dye reporting cell integrity.
Phenotypic Screening and In Vitro Toxicology: Applications of Far‐Red Fluorescing DNA-Specific Anthraquinones in Cell‐Based Assays
This application note demonstrates a number of applications of Fluorescein-/GFP-compatible dyes, in particular DRAQ5 and CyTRAK Orange. The particular spectral and chemical features of DRAQ5 and CyTRAK Orange mean they are compatible with aqueous media and fixatives. Conveniently, they are supplied in aqueous solution and can be applied as the final step in a sample preparation. They have become standard reagents for immunofluorescence microscopy, flow cytometry and high content imaging.
