"We use Kinetex 5 micron C18 columns in a synthetic lab for routine reaction monitoring. Most of the time it's a simple dip-dilute-and-shoot sample prep with no specific clean-up. The column survived well and gave excellent results, both in peak shape and sharpness. Some of our samples have very polar and chelating bisphosphonate groups, and the column handled them well. The 5 micron columns give 70~80% of the resolution power of what 2.7 micron columns offer, with significantly lower back pressure. They are well suited for a simple drop-in upgrade for older instruments that cannot handle the high pressure of small particle size columns. The new EVO columns give the same or better peak shape, with significantly wider usable pH range. We haven't used those for long enough to comment on their longevity. But the current column has survived over 500 tissue samples (blend and organic solvent precipitation) without notable degradation. Their customer care and technical support teams are very "

"Direct comparison of Supelco Discovery C18 (5 µm, 150x4.6mm) and Kinetex C18 (5 µm, 150 x 4.6mm): Analysed the same day on the same instrument (Agilent 1200), using the same sample and mobile phase: Instrument: Agilent 1200 Mobile phase: 20% acetonitrile in 5 mM acetate buffer pH 4.5 (isocratic) Sample: 20 µl of a 100 µg/ml solution (in-house peptide) Flow: 0.5 ml/min Temp: 50°C Retention time Supelco column: 18 minutes Kinetex column: 14 minutes (More retention on the Supelco column, maybe not so surprising) Tailing factor Supelco column: 1.8 Kinetex column: 1.4 (clearly better symmetry on the Kinetex column) Plate count Supelco column: 12.000 plates Kinetex column: 16.000 plates (I am a bit shocked that the Kinetex column gave so much better efficiency. I may have overloaded the Supelco column, giving a lower plate count and more asymmetry) Conclusion: The Kinetex and Supelco columns are probably comparable. The Kinetex column appears to have a better load."

"I developed the method and validated it with three different batches of 1.7 µm columns. It was performed on two different HPLC systems and three different analysts. Everything looked fantastic with the lowest USP resolution of the 10 peaks of 3.0 in all runs. The robustness testing showed that I could change pH and acetonitrile concentration in a wide range without any significant loss of resolution. The method is transferred to QC (in another country) without any problems. They buy a large number of columns for the process validation. Only a few works. Peaks are split or very broad. They record the backpressure (very good, 20% less than other competitors)."

It's a robust column, providing sharp peaks with stable retention times, which leads to a short HPLC-method (ca. 20 min). We could change our former 2.6 µm Kinetex column into this 5 µm column without loss of resolution or elongation of the method. The 2.6 µm column got dirty quite fast by small particles that weren't held back by the guard-system and had to be replaced. This is not a problem with the 5 µm column. It is flushed and cleaned easily and thus can be used for a longer time before it has to be replaced. "

"I used this Kinetex column in my pharmacokinetics study. After using this column in my high performance liquid chromatography coupled with electrochemical detection, I felt that this column had the better separation and good reproducibility and the sensitivity is good. However, when I did my study, I had some problems. Because my study is about pharmacokinetic with microdialysis technique, my samples include a lot of salts. I felt that the reservation is very important. I usually clean my column with 10% methanol first and then with 50% methanol. It usually took a lot of time to clean the column."

It is very important in the regions of Italy (Puglia and Basilicata), mostly in samples under seizure protocol because this column allows analyses which are fast and sensitive. Screening methods are ELISA and confirmatory analytical method is UHPLC with fluorescence detection with post-column derivatization with orto-phthalaldheide (OPA) and thiofluor. The legal limits are 100 mg/kg and 200 mg/kg in fish products. The confirmatory method is very fast (running time is 10 minutes and total time analysis is of 25 minutes). The sample is formed by 9 units, therefore fast analysis is very good."
