Perfect UPLC for method development

Average Rating 4.7

Application Area:

New born screening

The online sample preparation module with the quaternary pump is the best in the class of UPLC module . Significantly reduce the sample runtime and have best in class solvent management system . Pilot projects and method development is facilities by the 4 mobile phase line which is further given a consistent flow and mixing by the quaternary pump. Further more the advance hand tight ferrules is easy to handle and decreases the void volume significantly . We use tandem MS as primary detector and the signal to noise ratio for the lowest detectable concentration of analytes is noticeably better than any other module we have used so far . All in all the consistent results with accuracy of area under the curve even after 30 injection of known standard is best we get for the price range and even beyond .

High Salt Gradient Analysis of Post-Translational Modifications - Deamidation Monitoring

This application shows an easy deamidation monitoring using the Vanquish Flex UHPLC system with UV detection. Therapeutic proteins have a major role in today’s medical treatment for a wide array of diseases. The diversity and efficacy of biotherapeutics makes them ideally suited for the treatment of various diseases such as rheumatoid arthritis, multiple sclerosis, and several types of cancer. Due to the complexity of biotherapeutics, they are prone to post-translational modifications that have to be characterized. One of the most common non-enzymatic post-translational modifications in therapeutic proteins is the deamidation of asparagine residues. It has been reported that deamidation can lead to allergic reactions as well as a reduction of the half-life, pharmacological dynamic, stability, or even a loss of the biological function. Thus it is crucial to characterize and control deamidation products in biopharma processes to ensure a high quality product.

LC-UV-MS Peptide Mapping Development for Easy Transfer to LC-UV QA/QC

Peptide mapping is one of several routine methods to characterize biopharmaceutical proteins. For research environments, this technique, if combined with mass spectrometry (MS), is utilized for the characterization and confirmation of the primary sequence of monoclonal antibodies. In addition, peptide mapping can help to identify, localize, and quantitate post-translational modifications (PTMs). Peptide mapping methods are often developed and evaluated with combined UV and MS detection, to simplify the transfer to routine environments where UV detection is used alone. In high-throughput workflows, peptide mapping experiments are performed for antibody identity confirmation, PTM characterization, and stability studies. In this work, the separation of peptides obtained from a monoclonal antibody digest is demonstrated with a LC-UV-MS setup.

Reliable Results in Peptide Mapping Using the Vanquish Flex UHPLC System

Peptide mapping is one of the routine methods for biotherapeutics characterization. This technique, combined with mass spectrometry, is utilized in research environments for the determination of the primary sequence of an antibody or the identification of post-translational modifications. UV is often used as a unique detection tool, for instance in stability studies. The data interpretation of these experiments is based on retention times as qualitative and peak area as quantitative information. In this application note, the separation of peptides obtained from a bovine serum albumin (BSA) digest is demonstrated. The retention time and peak area precisions are evaluated for repeated injections.
