We received this column just over a month ago. For the first 100 injections everything was very good. Separation was wonderful. Analysis of a sample took 15 minutes. I thought that I received the best column which you can buy. I used a cleaning solution of 0.1 M Monosodium phosphate pH 3.0 and 20% methanol. A week later I used the column again but separation was awful. I tried to clean it with 0.5 SDS and acetonitrile gradient, but result was the same. Then I connected a 2 year old BioSep-SEC and result was good. So problem was in Yarra column. Phenomenex provide 45 days of return or exchange for their columns. I contact domestic distributor and explain situation. But until now, nothing has changed. Annoyingly that column for more than 1,000 Euro died so quickly.

This application note demonstrates the performance advantages of Yarra 1.8μm SEC-X150 Columns from Phenomenex for the analyses of biomolecules by aqueous size exclusion chromatography (SEC). The switching of mobile phases was used to simulate stress on the silica bed. Faster flow rates served to increase the backpressure to atypical levels and the stoppage of flow after each series of steps mimicked the completion of a series of samples. Column failure was not actually achieved after this study was completed, modeling the stable performance of Yarra SEC-X150 columns over longer lifetimes.

Size Exclusion Chromatography Method Optimization using the European Pharmacopoeia for Insulin Fibrils

Insulin is a peptide hormone which plays a central role in metabolism. In its monomer form, insulin can partially misfold, causing a cascade of non-native cross assembly of beta-pleated sheets or amyloid-like fibrils. A common method for analyzing insulin HMWP is size exclusion chromatography (SEC). However, the method requires a lengthy 35 minute run time which is time prohibitive for any laboratory with many samples to run and data points to generate. This application shows the method transfer of the Ph. Eur. method from a traditional, 10µm hydrophilic silica 300 x 7.8mm column to a high performance, 1.8µm hydrophilic silica 150 x 4.6mm column.
