• CYP3A4 + P450 Reductase + Cytochrome b5
• Control: Insect Cell Membrane Protein
• Buffer Solution: 0.5 M Potassium Phosphate (pH 7.4), filter sterilized
• Stop Solution: 0.5 M Tris Base, filter sterilized
• Cofactors: 1.3 mM NADP+, 66 mM MgCl2, and 66 mM Glucose 6-Phosphate
• Glucose 6-Phosphate Dehydrogenase: 40 units/mL in 5 mM Sodium Citrate Buffer (pH 7.5)
• BFC (7-Benzyloxy-trifluoromethylcoumarin)#: Fluorescence substrate, 4.3 mg
• KTZ (Ketoconazole): CYP3A4 selective positive control inhibitor, 0.008 mg
• HFC (7-Hydroxytrifluoromethylcoumarin): Metabolite standard (0.25 mM in 0.1 M Tris-pH 9.0)
