For many years, my core facility users in general and I, personally, have been using several BioStatus products, mainly DRAQ5 and DRAQ7. I have always found them effective and extremely easy to use. DRAQ7 has shown for many years to be a fantastic option for the detection and exclusion of dead cells. Quite recently, we have tested and used DRAQ7 DROP & GO for the detection and exclusion of dead cells from a very heterogeneous sample for isolating photoreceptors from adult zebrafish retinae for downstream RT-PCR. It was as easy as described in the name of the product "drop and go". Dead cells were clearly differentiated from the alive ones as can be easily observed in our recently published paper Glaviano et al. Application of DRAQ™ Dyes in Single Cell Transcriptomics and Genomics
DRAQ5™ and DRAQ7™ dyes provide accurate fluorescent labelling and assist in distinguishing cell status for single cell transcriptomics and genomic applications. One concern regarding the use of these DNA binding dyes is any unwanted effects on the first strand cDNA synthesis. However, studies in this white paper have demonstrated that DRAQ5™ and DRAQ7™ do not interfere with the synthesis process, indicating an accurate experimental outcome.
Channel- & Compensation-free Dead Cell Exclusion: N+1 Panel Design
Dead cell exclusion is a common requirement for flow cytometry using viability dyes like DAPI, PI and 7-AAD. Anew far-red fluorescing alternative DRAQ7™ has been robustly tested showing improved separation between negative and positive events. Conventional flow cytometry is limited by spectral "space" and the preference for bright chromophores to label antibodies to aid sensitivity that increases channel spillover. At its simplest, far-red DRAQ7 can benefit panel design where, for example, the complete absence of spillover into the FITC and R-PE detectors makes it an obvious replacement for 7-AAD in CD34+ cell enumeration.
