Application Area:

Analyse cell viability in compound screenings

I use DRAQ5 for cell staining to enable a precise and reliable quantification of the viability of my cells before and after the application of compounds at varying concentrations. It's very easy to use with no need to suspend the stock solution, and readouts of the nuclear stain are always crisp! I have never had any problems with the reproducibility of results, with clear and precise control values every single time. There is honestly nothing bad I can write about this product as I have never had any problems, and it's so simple to use. I do not personally purchase this product, but my team has never had any problems with ordering and delivery, with fast turnaround times.

Good for necrotic nuclei, pity that Draq5 and Draq9 are at the same wavelengths.

Average Rating 4.0

Application Area:

Visualize necrosis on glioblastoma primary spheroids with LSFM imaging

Glioblastoma spheroids of ~1mm in diameter are incubated with Draq7 for 16-20hrs before imaging. Draq7 is visualized in far red and stains dead nuclei, without discriminating cell death pattern. Easily combined with blue and green probes. Photobleaches after 6hrs.

Application Area:

Assess nuclei in 2D and 3D immunofluorescence

We use this product to look at nuclei in both 2D and 3D fixed immunofluorescence imaging. Very useful if you do not have access to a 405 laser and cannot use DAPI, as this uses the 647 channel.

DRAQ7 fluoresces in the far-red – with the same spectral properties that have made DRAQ5™ a reagent of choice in HCS.
DRAQ7™ only labels membrane-compromised dead, dying and apoptotic cells and gives clear discrimination between these and healthy cells in high content cytotoxicity and viability assays and in flow cytometry.

The far-red emission, longer wavelength than TOTO-3, permits better combination with other fluorescent probes. It is spectrally compatible with the commonly used orange/red mitochondrial health probes, and UV-excited vital dyes.

DRAQ7 can be used in RNAi knock-down and long-term, dynamic viability assays to report in vitro cytotoxicity and apoptosis.

Also, DRAQ7 efficiently counterstains fixed cell and tissue preparations. It offers the same differential nucl:cyto staining for dual compartment segmentation in translocation tracking and cell morphometrics for in vitro toxicology, that has made DRAQ5™ a favourite of HCS assay developers.

DRAQ7 is supplied as a convenient aqueous, ready-to-use solution. The 1 ml pack size (GBP 140.00 / ca. USD 220.00) is sufficient for at least 200 flow cytometry tests, 300-600 slides and up to 10,000 1536-MTP wells.

DRAQ7 in Cell Health Assays

One of the biggest costs in drug discovery and development has been failures of drug candidates after launch, due to unexpected or idiosyncratic toxicity. In this application note, learn how the DNA-binding viability probe, DRAQ7, is used in image-based and FC-based cell health assays to determine dead or dying cells.

White PapersLife Sciences

Real-Time Cell Health Monitoring, Cell-by-Cell, in 2D and 3D in the Far-Red

A reliable estimation of cell viability is central to assays for tumor cytotoxicity, apoptosis, and in vitro toxicology. DRAQ7™ is a membrane-impermeant DNA probe that enters leaky cells during apoptosis or necrosis to label the non-viable population. This white paper demonstrates the validation of DRAQ7 for use in long-term and real-time cell health assays, in combination with live cell-permeant DNA dyes for live/dead cell counting.

Application of DRAQ™ Dyes in Single Cell Transcriptomics and Genomics

DRAQ5™ and DRAQ7™ dyes provide accurate fluorescent labelling and assist in distinguishing cell status for single cell transcriptomics and genomic applications. One concern regarding the use of these DNA binding dyes is any unwanted effects on the first strand cDNA synthesis. However, studies in this white paper have demonstrated that DRAQ5™ and DRAQ7™ do not interfere with the synthesis process, indicating an accurate experimental outcome.

APOPTRAK™ in FC-Based Apoptosis Assays: Live Cell Marking and Cell Death Scoring

In apoptosis an ordered series of events leads to destruction of a single cell. The changes that occur can be assayed through DNA fragmentation, caspase activity (using permeant substrates e.g. FLICA™ probes), mitochondrial membrane potential (e.g. using TMRM), cytochrome C release, plasma membrane inversion (using Annexin V) and permeabilization being common choices. This application note demonstrates the use of APOPTRAK™ in the analysis of far-red fluorescent dye reporting cell integrity.

DRAQ7™: Identifying Membrane-Compromised Cells in Cell Health Assays

One of the biggest cost in drug discovery and development has been failures of drug candidates late in the process (or after launch) due to unexpected or idiosyncratic toxicity, not to mention the risk to patient health. Recently, cell health assays have been developed that use physiologically relevant cells and measure their health under different doses of a compound using a few simple parameters. DRAQ7TM has been validated for long-term, real-time, cell health assays and exhibits extremely low photo-bleaching. The use of DRAQ7TM in the identification of membrane-compromised cells is described in this application note.
