Epigenetic identification and targeting in clinical FFPE tissues is a crucial step for potential epigenetic therapeutics and personalized medicine therapies. FFPE is a standard method to archive and preserve medical tissue biopsy samples. These archives, kept in many hospitals world-wide, are a huge, rich and untapped sample resource for genetic research. Due to the cross-linking of proteins by formaldehyde, FFPE tissues present a particular challenge for ChIP analysis. In addition, the small size and delicate nature of the tissue, and the difficulty in extracting samples from paraffin, used to provide tissue support after preservation, can damage complex cellular complexes such as chromatin. ChIP assays from these samples have therefore proved time consuming and difficult.
Chromatrap® FFPE ChIP kit overcomes these issues and provides the following benefits:
- Provides greater flexibility and more IPs per sample
- Works across a range of FFPE samples, human and animal
- Available in 96-well plate or spin column format
- Works equally well with high and low abundant marks
- Provides sufficient DNA to perform library preps for ChIP-seq assays
Unlocking the Archive – Chromatrap® ChIP from Formalin Fixed Paraffin Embedded (FFPE) Tissue
The chromatin from preserved tissues from biopsies provides the field of epigenetics with a vast archive of information about histone modifications transcription factor occupation of gene promoters in a multitude of disease states. This application note describes how the Chromatrap® FFPE ChIP kit can aid extraction and analysis of chromatin from both human and animal tissue preserved using FFPE. This article highlights the benefits of Chromatrap® FFPE ChIP kit including efficient extraction, high yields of chromatin, shorter protocols and high throughput capability.

Chromatrap® FFPE: Efficient Analysis of Epigenetic Alterations in Cancer
Due to the cross-linking of proteins by formaldehyde, FFPE tissues present a particular challenge for ChIP analysis. Learn how the Chromatrap® FFPE ChIP kit can overcome these issues to provide sufficient DNA to perform library preps for ChIP-seq assays. Examples of chromatin extraction from human FFPE tumor chromatin, human breast FFPE tumor tissue and rat FFPE uterine tissue are presented.
Highly Efficient ChIP qPCR Assay Kit for FFPE Samples
In this application note, Chromatrap® introduces a new Formaldehyde Fixed Paraffin-Embedded (FFPE) ChIP-qPCR Kit which overcomes the widely acknowledged shortfalls of the technique. Available in 96-well or spin column formats the new Chromatrap® FFPE ChIP-qPCR kit not only provides greater flexibility and more IPs per sample but is also proven to work well across a range of human and animal FFPE samples. Requiring 10x less starting material compared to traditional methods, the Chromatrap® FFPE ChIP-qPCR kit produces 4x better pull down than even the best competitive kits. The new Chromatrap® FFPE ChIP kit works equally well with high or low abundant marks and provides valuable data where the transcription factor loci are already identified.
