Tricyclic Antidepressants (TCAs) are prescribed for the clinical treatment of mood disorders like depression and bipolar disorders and can also be used in long-term pain management therapy. They work to prevent the re-uptake of serotonin and norepinephrine in the brain. The monitoring of TCA levels in patients is important to the continued prescribing of the drug and overall determination of patient response. This application note describes the extraction of a range of opiates from human urine using ISOLUTE SLE+ supported liquid extraction plates (96-well) and columns with LC-MS/MS analysis.
This method describes the use of ISOLUTE SLE+ supported liquid extraction plates (96-well) and 1 mL sample volume columns to extract a range of opiates including opiate treatment analytes (methadone and its metabolite EDDP)) from human urine. This simplified and efficient extraction method has significant analyte recoveries ranging from 70-102% with LOQs as low as 500 pg/mL.

Extraction of Benzodiazepines From Human Urine Using ISOLUTE® SLE+ in Column Format Prior to GC-MS analysis

This application note describes the extraction of a range of benzodiazepines from human urine using ISOLUTE SLE+ supported liquid extraction columns prior to derivatization and GC-MS analysis. Benzodiazepines are psychoactive drugs used to treat various conditions including anxiety, insomnia, seizures and alcohol withdrawal. This application note details an optimized extraction protocol for the use of ISOLUTE SLE+ supported liquid 1 mL sample volume columns to extract a range of benzodiazepines from human urine. This simplified and efficient extraction method has analyte recoveries from 90% with LOQs as low as 40 ng/mL. The application has also been performed with halogenated and non-halogenated extraction solvents in the interests of providing for green chemistry.

Extraction of 11-nor-9-carboxy-Δ9-tetrahydrocannanbinol from Urine Using ISOLUTE® SLE+

This application note describes the extraction of THC and metabolites from urine using ISOLUTE SLE+ columns or plates. Globally, cannabis is one of the most widely used illicit drugs. The naturally occurring cannabinoids found in hemp species bind to receptors in the brain and cause sensations of relaxation and calm. Widespread legislation against the use of cannabis has led to the necessity for rapid and reliable methods for the analysis and quantitation of cannabinoids and metabolites. The most prevalent marker in biological samples taken from cannabis abusers is 11-nor-9-carboxy-Δ9-THC. This study demonstrates a supported liquid extraction procedure for 11-nor-9-carboxy-Δ9-THC.

Read this application note to learn more about the extraction method for 22 different drugs in urine which are typically screened for pain management panels using ISOLUTE SLE+ fixed well plates. The use of schedule I drugs for patient pain management therapy warrants constant monitoring of therapeutic levels in the patient. Screening patient urine samples for the free drugs is complicated by the metabolism process which converts the free drug to the ß-glucuronide form. Patient urine samples can be enzymatically hydrolyzed and extracted to detect the drugs using Supported Liquid Extraction

Effective Strategies for Phospholipid Removal using Supported Liquid Extraction (SLE) with LC-MS/MS Analysis

This poster compares phospholipid removal using a wide variety of solvent combinations, pH control and polar extraction solvents on supported liquid extraction (SLE) plates. It was shown that the strong retention characteristics of phospholipids and subsequent co-elution was overcome using SLE. Phospholipids were observed at a low level with a variety of pH pre-treatment/extraction solvent combinations.
