Gene Fragments

I have used several other companies for gene fragments in the past, and one issue I frequently encountered was minimum order requirements. I would often have to purchase more fragments than I actually needed, which made the overall order unnecessarily expensive. Other companies that avoid these minimums can also be quite costly. Most importantly, I could order the number of fragments I actually needed without being forced to spend significantly more. I was also really impressed by the cloning efficiency. I have rarely seen such consistent colony growth and colony sizes. Using the same cloning and miniprep protocols I normally use, I consistently obtained high plasmid yields, often around 500 ng/µL, with A260/280 ratios around 1.8 across my samples. The resulting plasmids have also performed very well for AAV production, yielding high viral titers. The protocols on my end have not changed; the main difference was using Twist Bioscience for the DNA synthesis. Based on my experience, I would recommend Twist to both people who are learning molecular cloning and experienced researchers looking for a reliable and convenient option. Twist is great, reasonably priced and fast. However, I think they should improve pricing on fragments cloned in a plasmid for long sequences/large protein expression vectors and allow for the synthesis of more complex DNA. I often have to adjust my sequences several times before their algorithm accepts it.
