The results were good, but DraQ5 was too expensive, its utilization was limited.

"The product worked well. Our main problem with these stains is that they cannot be used in any microscope, e.g. Draq5 can be used in a confocal laser microscope, but cannot be used in another, simpler microscope, so we have to purchase different stains for different microscopes. In addition, DraQ5 is more expensive than other similar stains."

You may wish to explore the use of CyTRAK Orange which is excited optimally at 510 nm (488 ex. compatible) and with peak emission at 615 nm (“rhodamine" channel). An easy-to-use DNA dye suitable for tissue clearing of mouse brain samples

"The DraQ5 fluorescent probe is supplied as a water-soluble concentrated solution (0.5mM or 5mM) that can be diluted depending on the desired application. Given its high affinity for double-stranded DNA, DraQ5 can be used as a nuclear counterstain in immunohistochemistry studies upon short incubation periods of time, as an alternative to blue-fluorescent DNA-binding probes. However, the broad excitation and emission peaks must be taken into account if a combination with other fluorophores is desired. In our experience, it is also compatible with aqueous-based tissue clearing techniques, rendering a uniform labelling across entire mouse brain hemispheres (55-days-old) that could be imaged by light-sheet fluorescence microscopy. To achieve this, a simple incubation at 37⁰C/60rpm for 12h with a DraQ5 diluted solution (2.5uM) and a 12-hour washing step at 4⁰C to remove excess were incorporated in the tissue clearing procedure."

Dear Zi, It would be helpful to understand what the issue was in more detail. If staining is not stable over time that can be the case in live cells, where after several hours a re-distribution can occur in some cell types, mostly towards lipophilic structures in the cytoplasm (Martin, 2005). If the staining is not stable in fixed cells then this may be due to washing post-staining, too low concentration, or not removing surfactants used in prior cell permeabilisation procedures.

Far red cell nucleus stain

"DRAQ5 is generally easy to use. The spectra are in long wavelength that can be paired with many other fluorophores and thus our choice of nucleus dye for confocal imaging. However, the performance is not very stable, sometime it can be very dim and unspecific staining outside the nucleus can be seen. The product (from Thermo Fisher) is very expensive. It will be good if the price can be more reasonable."

Dear Yuning, thank you for detailed review of DRAQ5. Without seeing examples of the issues you raise it is difficult to give further technical advice. The occasional cytoplasmic staining you describe is often a symptom of understaining or washout due to residual surfactant left over from a prior washing step. Regarding cost, there are a number of other carefully selected suppliers you can choose from and we can advise on the most competitive of these or alternatively order direct from us, the source!

"DRAQ5 is very easy to use. You resuspend cells in PBS, stain cells in dark for 1-3 minutes, then you can image cells directly (no rinsing is needed). The product is kind of expensive, but it is worth it. The dye can be excited by either 488 or 639 nm laser, but emits in far red, so it won't interfere with GFP."

"The DRAQ5 reagent is very easy to use. I have used it together with DAPI for counterstaining the cellular nuclei in various cell types like SHSY-5Y, SKN-SH, U2OS and HeLa. The reagent is robust and the staining is perfectly reproducible. I had used the probe in a variety of application like Fluorescent Microscopy, FISH and combined FISH with ICC. The probe lifetime is commendable even after multiple exposures. You can find images with application of DRAQ5 in my publication here - Chanda, K., Jana, N.R. & Mukhopadhyay, D. You make a good point too: it is possible to view DRAQ5-positive cells with transmission light microscopy - the nuclei are indeed deep blue (as well as far-red fluorescent)! It’s a convenient way to test for functionality and sample quality (for example prior to FACS) and might even allow for a new breed of assays.

The product code is DR05500 and, having reviewed your protocol, would perhaps allow you to skip the pre-dilution step entirely e.g. 4 µl into a final volume of 100 µl would give a final concentration of 20 µM. Perhaps this might assist you better?”

"DRAQ5 is an easy-to-use product that is ready straight from the bottle and just have to be diluted to a working concentration. As with almost all dyes and antibodies for immunofluorescence, it has to be tested for the ideal concentration but the product came with suggested working solution concentrations that proved to be very useful as I had to do just a couple of tests to establish the working conditions. The dye is very bright and long-lasting, and the reagent itself lasts for long time at 4 degrees C."

Decent nuclear staining in far red channel.

"If there is the need to visualize fixed nuclei by confocal microscopy in the far red, then Draq5 is the stain of choice. Of course it is not as bright as DAPI and probably the "gain" and "laser power" should be considerably increased. Also some leakage of staining in cytoplasm could be observed, but still it is very reliable for staining. For the best results, dilution 1:500 in 0.1M PBS (alternatively with 0.3% Triton X) should be used."

Thank you very much for the valuable feedback and product review on DRAQ5. The observation about the use of formic acid is useful to know. Perhaps it may be worthwhile to attempt staining with DRAQ5 as the final step after all other steps are complete, including the formic acid treatment. It is unusual to see staining of other structures outside of the nucleus with DRAQ5 but may be a consequence of the treatments of the sample. DRAQ5 does have, like all cell permeant drug-like small molecules, a propensity for binding to lipophilic structures in addition to the specific target, in its case dsDNA, but this is usually only seen in cases of understaining, paradoxically. We are always ready to answer these questions through our tech support service as they occur in real-time in the bench experiments.

"We used DRAQ5 because of its far-red fluorescence and suitability for live/fixed-cell imaging. In general, it suits our needs as we can't find a commercial alternative out there. It is easy to use, but sometimes the fluorescence can be very dim and needs some adjustment to make it visible. We bought it from ThermoFisher/Invitrogen so it is quite expensive."

Thank you for your review of DRAQ5. DRAQ5 does have a useful weak secondary staining of the cytoplasm which HCS labs use to segment the cell envelope, by increasing the gain. An unwanted inversion staining (“hollow” nuclei) is a consequence of too low a concentration of DRAQ5. We strongly recommend 5 uM for fixed cells and 5-10 uM for live cells. We would be very happy to discuss this and troubleshoot the issue to try to resolve it.

The initial review posting elicited an immediate technical support response. After successful testing of both fresh DRAQ5 and DRAQfx (for comparison) in the customer’s lab the issue was isolated to the presence of 0.3% Triton X-100 during the final counterstaining step. This was remedied by an intermediate wash with PBS. This learning will be reflected in our products’ technical data sheets for fixed cell immunofluorescence protocols.

The presence of precipitates in the vial is most unusual and could only be a result of contamination or freezing (accidental, most usually). We would like to have explored this with you at the time you saw this. Your comment about smaller quantities for fixed cell staining is well made - we have developed a product exactly of this nature - please see DRAQfx which is specifically for fixed cells, provided in a convenient dropper bottle.

Thank you Anirban! Yes, DRAQ5 will also work for fixed cell fluorescence microscopy. You can use it prior to fixation, but more usually it is used AFTER fixation of the sample. We also have a new convenience alternative - DRAQfx FIX & GO - which has been specifically designed for the latter use and may exhibit less cytoplasmic background, though typically this is extremely low.

This reagent is very easy to use, extremely reliable and efficient as it quickly enters cells. It displays a very bright fluorescence with only a very small amount. Support from BioStatus is outstanding. It is also very easy to use and does not need to be washed."

"The product works very well and can be re-used. I used it as an alternative to DAPI and Hoechst, to stain cellular DNA within the nucleus, because the CLSM system we use does not have a 405 nm laser line. The main problem of this product is that it is very expensive when compared to other stains such as the ones mentioned above. Indeed for such application I would not recommend it. However, it is ideal for live cell imaging experiments since it is cell permeable and the 633 laser used for its excitation is much less phototoxic as compared to the 405 one."

DRAQ5 is second generation DNA probe that enables live or fixed cell counterstaining and avoids UV exposure as you rightly observe, plus with much lower photo-bleaching and remarkable shelf stability. Being red-excited it also enables use of the new violet-excited chromophores to extend channel capacity. Bearing these significant benefits in mind, the cost per slide of around 30 eurocents (based on 5uM, 200ul per slide, 200ul pack list price) would seem to be not unreasonable!

Excellent DNA dye - we use for single staining in antiparasitic high content assays

"Ideal for single-staining high content assays, DRAQ5 enables clear visualization of cell nucleus and gives a smoother background signal at cell cytoplasm - which is enough for image processing softwares to delimitate and segment cellular structures properly. Moreover, the stain stability enables material storage for longer period than other conventional dyes (we have imaged 384-well plates after 4 weeks post-staining and the fluorescence signal was sufficiently high); also there is no need for dye washout after staining the microplates (less pipetting stesp). We recommend DRAQ5 in single-staning high content screening for assessing compound activity against intracellular trypanosomatids (Trypanosoma cruzi and Leishmania spp.) - the dye marks both DNA material from intracellular parasites and host cells - and providing a faint signal in the cytoplasm - turning it easy to segment and quantify the intracellular parasites."

"DRAQ5 is a simple, easy-to-use product. Either used on live-primary or fixed cells a simple 5-10 minute incubation followed by washing steps produced consistent, repetitive results. Its fluorescent excitation/emission spectra are very well suited to combine with other fluorophores without channel bleeding or crosstalk. The particular spectral characteristics of DRAQ5 make it also easy to work with in fluorospectrometry-based experiments. We have also used DRAQ5 combined with metabolic biomarkers in solution without observing interference with cellular metabolic uptake in short-time uptake experiments, this allowed us not only to reduce experimental time, but also helped us reduce cell manipulation."

"DRAQ5 is a useful reagent for analysing the cell cycle status of live/dead cells by flow cytometry. For our purposes, cells were first killed by fixation in 70% Ethanol (because we were co-staining for other antigens) and eventually resuspended in buffer containing 2ul/ml DRAQ5 allowing 10 mins prior to flow cytometric analysis. Lovely DNA profiles were obtained with no compensation issues with the other fluorochrome (PE) used in the study. Never attempted on live cells but we understand from other colleagues in the department that this is possible. Care must be taken when handling as this is a toxic substance."

This is important information – we do not recommend DRAQ5 for long-term exposure to live cells as it, like ALL high affinity DNA binders and intercalators, will have some kind of impact on cell health. There are exceptions especially with terminally-differentiated cells as observed with HUVEC, macrophages and BaF3. However, short-term exposure and use in live-endpoint assays has been a hallmark of the product’s success in drug discovery and flow cytometry applications, in particular the loss of cytoplasmic GFP signals when cells are partially permeabilized by fixation with formaldehyde and the unspecific loss of cells due to RBC lysis and associated washes, respectively.

"The product is very easy to use for microscopy, in few minutes your sample is colored. I will recommend to researchers to use it if you have laser or filters for it. It is reliable and results are reproducible. We perform studies on melanoma and retina cell culture, exposed to different stress conditions and we follow their morphology after treatment."

"We used DRAQ5 to label the nuclei of live adult rat ventricular myocytes (see Weeks et. al. β-Adrenergic Stimulation Induces Histone Deacetylase 5 (HDAC5) Nuclear Accumulation in Cardiomyocytes by B55α-PP2A-Mediated Dephosphorylation. Journal of the American Heart Association. 2017 6(4)). We labelled the cells with 1 uM DRAQ5 for 10 minutes prior to imaging, which was lower than the recommended concentration, but it fluoresced nicely."

  • Application Area:To analyze nuclei in the cultured isolated bovine chondrocytes

"The dye was highly effective as even at very diluted concentrations produced excellent results. The confocal images obtained from these chondrocytes whose nuclei were labelled with DRAQ5 produced high quality and reproducible results. I had used this dye to count the number of cells in clusters because in my culture experiments the cell membranes were frequently touching and therefore thresholding of individual cells was impossible. In order to count number of cells in clusters I counted number of nuclei and DRAQ5 gave me highly reliable results as I could count the number of nuclei in 3D by using Volocity (a 3D image analysis software). It is quite an expensive dye but as the very diluted concentrations are very effective therefore, justifies the cost. Moreover, it was easy to use and highly penetrable and required very little time to label nuclei efficiently."

A very useful product for live cell imaging.

"The dye gives strong staining and could we washed away even from live cells. Nuclei of some cell lines e.g. PC3 could not be stained (the dye does not enter the cells) others e.g H1299 contain strange dotty staining in the cytosol in addition to the nuclear staining. Too high concentration causes artificial condensation of chromatin."

"This product allowed us to perform screens of compounds on cells that could not be assayed using traditional cell proliferation assays like Alamar Blue or MTT. The dye allows rapid quantification of cell numbers in a 96-well plate format. Assay of wells with multiple cell doses generated nice curves that gave us confidence in the quantitative ability of the assay over a wide dynamic range of cell numbers."

"Product (DRAQ5) nucleic acid stain works exceptionally well, and keeps common microscope filter channels open since it's a far red stain. Little to no background, easy to use and store. Penetrates deep into thick tissues so you don't observe staining concentration gradients as with other nuclear stains. Perfect for use when imaging 3D tissue constructs with confocal - but also works well for 2D monolayer applications where UV light isn't an option (ex: PMMA microfluidic devices which block UV)."

"If you are looking for a dye for in vivo staining, DRAQ5 is the best. Even with very little amount of a stock of the product, like 1ul, you can observe bright nuclei immediately. Staining genomic DNA specifically you could capture where DNAs are distributed in nuclei. After decorating your nuclei with it, cells are still happy living healthy. I will keep using the product for my future projects."

  • Application Area:Confocal imaging of parasites

"Excellent product. I had earlier used Giemsa for staining the parasite in macrophages. I work on a protozoan parasite and once I worked with DRAQ5 phew I got my paper accepted in Autophagy. Would recommend to all."

Nice images with confocal laser scanning microscopy

"Worked with diluted stock solution in PBS (better handling and accuracy). The applied incubation time, temperature and concentration were important for good reproducibility. On permeabilized cells the nucleus had better signal and stained clearly. On the non-permeabilized cells, the cytosol was stained also."
