I have used your NanoSight LM 20 for the characterization of exosomes. It provided excellent insights into the size and concentration of exosomes, and it was very easy to use. The results were highly specific and logical. Recently, I noticed that you have launched an advanced version, NanoSight Pro. I am interested in purchasing it to further advance my research on bionanoparticles/exosomes. This is truly an excellent instrument for determining the size and concentration of exosomes and other bionanoparticles.

Works well for sizing and quantifying LNPs and adenovirus

I've been using the Nanosight Pro for several months now to characterize LNPs and adenovirus both around 100 nm. Overall, my experience has been positive. There are definitely some challenges that come up and require experience. I've contacted support multiple times which have been good experiences. Pros: works great for quantifying and sizing my particles. Results can be reproducible. The video is useful and allowed us to see some unusual aggregate chains. Running samples is straight forward. Cons: Running samples can be problematic if unaware of potential problems. Air bubbles are the biggest problem. These can be very difficult to remove sometimes and must be checked for after loading a sample. Thorough washing between samples can be required depending on the sample. The fluorescence capability is nice but so far particle brightness seems lacking. Particle bleaching was also observed. You can reverse the flow to reduce bleaching but I have not tried it. Other thoughts: I have found some LNPs require washing with detergent after a few samples because (I suspect) lipids are sticking to the plastic and degrading the image sharpness. The concentration (particles/ml) also needs to be optimized for your sample to get an accurate count and minimize carryover between samples.

The software is not complicated; it saves time for training the new users and running the experiments. It has autofocus and a camera that is easy and convenient. Also, the data was precise and accurate.

We purchased this instrument for our core laboratory. It will be utilized by several investigators for viral and extracellular vesicle research. There is not another instrument on campus that would provide the data that this does. It is easy to use and importantly automate for walk away use. The training was outstanding and the instrument is easy to care for.
