A great column for small molecules with ionizable groups.
Analysis of small molecules
These columns can be used over a wider range of pH, which makes them much more versatile than the classical silica-based columns. Furthermore, they come in a brad range of bonded phases, particle sizes, pore sizes and dimensions. i found the Gemini SAX particularly useful because I can combine both the ionic strength and the pH of the mobile phase, to impact the selectivity, thus achieving optimal resolution of all my analytes with shorter run times.

I have worked with Phenomenex Gemini C18 and Phenomenex Phenyl C6 and obtained very good separation of compounds with aromatic rings, namely phenolics in plant materials (mobile phase of 01.% formic acid + acetonitrile). Phenyl C6 is better if you aim a good selectivity between isomers.

Rapid LC-MS/MS method for monitoring bio-relevant levels of per- and polyfluoroalkyl substances (PFAS) in serum
In this application note, a quantitative workflow for the analysis of 22 per- and polyfluoroalkyl substances (PFAS) in serum was developed using a Gemini™ 3µm C18 column coupled with a SCIEX QTRAP® 6500+system. This targeted screening workflow provides a fast analytical method capable of accurately quantifying sub-nanogram per mL levels of PFAS in the human body.
Per- and polyfluoroalkyl substances are pervasive compounds used in a variety of industrial applications and found in a wide range of consumer products such as cookware, stain repellent, flame-retardant, and coatings. PFAS are considered environmental factors due to their persistent and bio accumulating nature. Bioaccumulation of PFAS in the human body resulting from environmental exposure is a growing public health concern. Recent studies have linked PFAS exposure to adverse health outcomes including:
- childhood health complications
- reduced kidney function
- thyroid disease
- hormone suppression
- decreased fertility
- increased cholesterol levels
- diabetes
Reducing PFAS interferences during human plasma and serum analysis with accurate mass spectrometry
In this application note, Phenomenex presents experimental data on the reduction of per- and polyfluorinated (PFAS) interferences in human plasma and serum analysis. PFAS are well known anthropogenic contaminants and are detected worldwide in most environmental media, including air, water and soil. Widespread human exposure has resulted in nearly ubiquitous detection of PFAS in human serum. In order to identify the potential health risks PFAS present to humans, accurate and robust analytical methods are required.
In this analysis, a Gemini™ C18 column coupled with a high resolution, high mass accuracy mass spectrometer was used to mass resolve perfluorooctane sulfonate (PFOS) and perfluorohexane sulfonate (PFHxS) from serum interferences. The EPA 533 PFAS list was selected as the target analytes. The interferences were characterized using their fragmentation patterns and through comparison with MS/MS libraries. Finally, the serum interferences were chromatographically resolved using a 6.5 min gradient.

A Fast and Effective Approach to Analyzing Hormones in Drinking Water Using SPE and LC/MS/MS
This application note illustrates a method for steroid hormone analysis from drinking water using an optimized LC/MS/MS method. Analysis time is reduced from 50 minutes down to about 15 minutes while maintaining excellent linearity (0.99 R2 value or greater for each compound) and low minimum reporting level MRLs. The method presented uses a Gemini® 3 μm NX-C18 HPLC column, which delivers stability at pH 1-12. This improved method should greatly increase lab efficiency and productivity.

This application note illustrates a method simultaneous analysis of nicotine, two of its major metabolites, cotinine and 3-hydroxycotinine, as well as nornicotine and anabasine from human urine samples. The method described uses Strata™-X-C solid phase extraction (SPE) cartridges for sample clean-up and concentration, followed by fast (<6 min) LC/MS/MS analysis using a Gemini® NX-C18 column.
HPLC-UV Analysis of Vitamin C from Tablets and Soft-Gels
The verification of label claim data in nutraceutical formulations has come under scrutiny by the FDA. Manufacturers and contract testing labs alike are looking for accurate and scientifically valid methods that are suitable for use with different formulations. There were two primary goals of this project: firstly, to determine an accurate and reproducible means of analyzing Vitamin C and secondly, to demonstrate the suitability for analysis of two commercially available formulations of Vitamin C.
