Reproducible results, easy to use and maintain.
Sample preparation
I used the Bioruptor Pico to prepare sonicated chromatin samples for ChIP assay. I have also used to sonicated DNA for MEDIP and MEDIP-seq. In addition, I use the Bioruptor Pico to sonicate protein lysates for WB and IP.

Amazing!
ChIP
Diagenode is a great company. We recently started using the Biorupter Pico in the lab and it's the best machine - so consistent and so user friendly. It's my lab child. I use the ChIP kits as well, and it's wonderful. All controls work, robust and really reproducible results.

Highly recommended biorupter.
Chromatin immunoprecipitation
The setup and instruction manual are easy to follow. With a 30s on 30s off for ten cycles I can obtain excellent shearing of my chromatin into 200bp fragments.

Stopped working within 6 Mo, horrible customer service.
Sonication of crosslink cell lysate
Poor customer service, make you jump though many hoops to prove their machine's are not working and even then wont believe their customers. The pico performance was never as good as they claimed and dropped of to nothing with in 6 months.

Best sonicator I've ever used.
Digest DNA for Chromatin Immunoprecipitation
Once setup the unit was easy to use and very reproducible. Once we got past the learning curve of setting up, the unit was the easiest I've ever used.

Consistent, reproducible results, even with a low number of cycles.
Sonicate DNA for library sequencing preparation
The Bioruptor Pico is a very well-manufactured piece of equipment and has so far worked flawlessly for sonicating DNA for library sequencing. It was very easy to un-package, setup, and run, and maintenance is also quite easy as long as you have a good source of lots of distilled water that is not de-ionized. Lastly, it is impressive that comparable results can be achieved with a lower number cycles than on Pico's larger sibling.

great results
ChIP-seq in tissues and cell lines
It is easy to use and saves more time while shearing DNA.
Review Date: 29 Jun 2016 | Diagenode
The Bioruptor® Pico is the new next-generation all-in-one shearing device, highly optimized for DNA shearing for next-generation sequencing, chromatin shearing for ChIP-seq applications, RNA shearing, protein extraction, and cell/tissue lysis.
The Pico is the shearing device of choice for sequencing applications, providing optimal yields, lengths, and consistency and excels in shearing different fragment size ranges, which are frequently required for downstream applications (e.g. bridge amplification) for sequencing. Moreover, the Pico provides flexibility in other applications saving the need to buy additional equipment. Unlike competing systems, the Pico has high throughput capabilities and can process up to 12 samples at once.
The only ultrasonicator you’ll ever need, optimized for:
- Next-Generation Sequencing (5 - 100 µl)
- Chromatin shearing (10 µl - 2 ml)
- RNA shearing
- Protein extraction
- Tissue and cell lysis
- Simultaneous sonication of 12 samples
- Small and light
Application of the Bioruptor® Pico for Shearing Chromatin Fragments for ChIP-seq from Human Tumor Tissue
This application note illustrates how the Bioruptor® Pico (Diagenode) can process various human tumor tissues for ChIP-seq analyses of transcription factors as well as epigenetic marks. Chromatin must be optimally sheared prior to immunoprecipitation and subsequent library preparation for Illumina sequencing.
Best Workflow Practices for ChIP-seq Analysis with Small Samples
Combined chromatin immunoprecipitation and next-generation sequencing (ChIP-seq) has become the gold standard to investigate genome-wide epigenetic profiles, however this method typically requires a large number of cells. This application note describes a method for carrying out ChIP-seq with a small numbers of cells, using the True MicroChIP Kit and the MicroPlex Library Preparation™ Kit from Diagenode and Advanced Analytical Technologies, Inc. (AATI). This enables reliable analysis of rare or difficult-to-obtain cell types.
Evaluation of Bioruptor in Shotgun Library Preparation for Massively Parallel Sequencing on Illumina Genome Analyzer II
The Illumina Genome Analyzer II (GA-II) performs massively parallel short-read DNA sequencing, with the potential to generate up to 10 gigabases in a single run. The technology provides researchers with a significantly cheaper alternative to conventional sequencing by capillary electrophoresis. A consistent and reliable method of DNA fragmentation is essential in Illumina library construction. This application note evaluates and compares two types of fragmentation methods: nebulization and shearing using the Bioruptor.
Bioruptor System from Diagenode: Designed Exclusively for Fast & Accurate DNA Shearing
The Bioruptor® Pico is the DNA shearing device of choice for sequencing applications, providing optimal yields, lengths, and consistency. This poster demonstrates its performance compared to that of a competitor. The Bioruptor® Pico can be easily programmed to modify duration of sonication for optimal fragmentation and produces simultaneous sonication of 12 samples, making it a cost effective solution.
Chromatin Preparation From Tissues for Chromatin Immunoprecipitation Using Diagenode’s Chromatin Shearing Optimization Kit - Low SDS and Bioruptor®
This application note describes the chromatin preparation from fresh or frozen tissues. The isolated chromatin can be used for chromatin immunoprecipitation assays using Diagenode’s iDeal ChIP-seq kit (C01010051). The following protocol is optimized for 30-40 mg of tissue allowing up to 18 ChIP samples (1.5-2 mg of tissue per sample). However, the exact amount of tissue needed for ChIP may vary depending on protein abundance, antibody affinity etc. and should be determined for each tissue type.
Transcription Factor Binding Studies with IP-Star® Compact Automated System from Diagenode
ChIP experiments require long and multi-step protocols, and the risk of introducing inconsistency is very high. The goal of this experiment was to confirm the findings from ChIP sequencing by using an automated ChIP technology in order to reduce all inconsistency introduced by the pipetting variations and human variability. This application note presents high sensitivity automated ChIP experiments to study transcriptional regulation and siRNA technology. The study used the Bioruptor UCD-300-TO with associated reagents for chromatin preparation and the SX-8G IP-Star® Compact Automated System with associated reagents for ChIP and an ETO antibody for the immunoprecipitation step.
