Transcription Factor Binding Studies with IP-Star® Compact Automated System from Diagenode

29 Oct 2014

ChIP experiments require long and multi-step protocols, and the risk of introducing inconsistency is very high. The goal of this experiment was to confirm the findings from ChIP sequencing by using an automated ChIP technology in order to reduce all inconsistency introduced by the pipetting variations and human variability. This application note presents high sensitivity automated ChIP experiments to study transcriptional regulation and siRNA technology. The study used the Bioruptor UCD-300-TO with associated reagents for chromatin preparation and the SX-8G IP-Star® Compact Automated System with associated reagents for ChIP and an ETO antibody for the immunoprecipitation step.

SX-8G IP-Star® Compact Automated System

Diagenode

Chromatin immunoprecipitation and DNA methylation assays followed by next generation sequencing is a powerful technique for generating genome-wide profiles of epigenetic modifications and transcription factor binding sites. The SX-8G IP-Star® Compact Automated System provides the flexibility needed to change multiple parameters in the protocols to achieve optimal experimental conditions. Samples generated on the system are compatible with popular downstream applications such as QPCR, microarrays and next generation sequencing platforms. In addition to automated ChIP and DNA methylation protocols, this system contains a wide variety of protocols including next generation sequencing library preparation and sample purification. Easy-to-use user interface for protocol optimization Magnetic-bead technology for great reproducibility Reduced "hand on time" to just only 15 minutes Easy to use bench-top liquid handling system Process up to 16 samples in parallel True "walk-away" automation Flexibility to adapt your experimental condition

Bioruptor® Pico

Diagenode

The Bioruptor® Pico is the new next-generation all-in-one shearing device, highly optimized for DNA shearing for next-generation sequencing, chromatin shearing for ChIP-seq applications, RNA shearing, protein extraction, and cell/tissue lysis. The Pico is the shearing device of choice for sequencing applications, providing optimal yields, lengths, and consistency and excels in shearing different fragment size ranges, which are frequently required for downstream applications (e.g. bridge amplification) for sequencing. Moreover, the Pico provides flexibility in other applications saving the need to buy additional equipment. Unlike competing systems, the Pico has high throughput capabilities and can process up to 12 samples at once. The only ultrasonicator you’ll ever need, optimized for: Next-Generation Sequencing (5 - 100 µl) Chromatin shearing (10 µl - 2 ml) RNA shearing Protein extraction Tissue and cell lysis Simultaneous sonication of 12 samples Small and light
