As a long-time user of a competitor's product, we switched to this Twist product very late. We were extremely surprised how good and above all stable the very good results were with repeated use. From the order to the results, everything runs smoothly. The enrichment works so evenly that we have expanded our analyses. We had previously tried many software for analyzing CNVs and were never satisfied with the results because we were used to CNV results from microarrays. With Twist exome enrichment, for the first time we were so satisfied with the CNV results that we stopped microarray analysis. That saved us a lot of money and a lot of work in the laboratory. The sequencing libraries are very reliable and very consistent in quality over time. The Library QC never has anything to complain about. It is very positive that the result corresponds very well to the performance described. The promised target is satisfactorily covered to over 99.5%.

The Twist Human Core Exome kit produces exceptionally high data quality, consistently meeting our threshold cut-off of 98% coverage at 20X depth. Coverage over some of the trickier GC rich/ repetitive regions of the exome is better than competitor products we have previously used, so requires less confirmatory or 'infill' testing by other methods (e.g., Sanger). The introduction of the Universal Adapter System part of the kit has improved our data recovery hugely, giving 10x less undetermined reads. The kits give good value for money for NHS diagnostics and now that kits are modular we have less wastage of excess reagents. Kits are easy to use given the complexity of library preparation and are easily automatable on a variety of liquid handling platforms. Customer service and technical support at Twist is exemplary, with detailed support given for troubleshooting and advice given well ahead of time to any changes in supply of reagents (e.g., due to COVID).
