Bioprocess Internship

The NC202 was used extensively throughout our summer internship programme at Extracellular. This allowed bioprocessing interns to effectively monitor the growth and health of QM7 cells on a daily basis. This piece of equipment was excellent for users new to the field, providing a user friendly interface with efficient, quick and reliable data.

Engineered CHO cell culture characterisation

The instrument was used to monitor CHO cell cultures in an academic setting. The cells often expressed fluorescent proteins as part of our cell experimental workflow. We speculated that there was a possibility of interference between fluorescent protein expression (especially mOrange) and the viability measurements returned by the NC-202 Nucleocounter. We were very pleased to see that fluorescent protein expression had no impact on the accuracy of the viability measurements. Moreover, we had the opportunity to benchmark the NC-202 with manual counting and another automated cell counter from a different provider. The accuracy in quantifying cell density and viability of all our CHO cell cultures was comparable across all methods. Importantly, the ease of use of use of the NC-202 exceeded that of the other two methods, as it did not require dilution or further sample handling. Overall, we were very impressed by the NC-202's performance! I also want to mention that engagement from the ChemoMetec team (commercial and technical) was exceptional! Very rapid response times and genuine interest in assisting in my group's work. Thank you!

Reliable results, no matter the sample!

At Infinitopes, precision really matters to us — not only in our target discovery but also in cell counting which is imperial for producing our vaccine vectors and their quality assessments, to testing how our epitopes trigger immune responses. Accurate ad consistent cell counts are key to keeping our research reliable and our data solid. We have used the NC-202 across a range of cell types — HEK293, A549, A375, CaSki, SHP77, and various primary cells — and it’s handled them all with no problems. For adherent cell lines, once they’re trypsinised, counting is straightforward. But even when we skip trypsinisation and leave cells in their natural, sometimes clumpy state, the NC-202 still delivers. Its lysing solution has been a game-changer for getting accurate counts where trypsinisation was not possible for our antigen discovery experiments or when manual methods were to laborious. Compared to the multiple other machines we’ve trialed, the NC-202 stand out first. Its software does an excellent job of recognising and counting cell clumps, and the lysing solution really solves any aggregation issues. We’ve also had great results with PBMCs, which are notoriously tricky due to their size and shape differences. The dedicated PBMC protocol has given us counts that match manual accuracy and since the system is 21 CFR Part 11 compliant it means that is suitable for our clinical trial work that is underway. Overall, the NC-202 has proven incredibly versatile and dependable. Whatever type of sample we’ve thrown at it, it’s delivered accurate, reproducible results - usually you can't have both and there's a trade-off in between the two, but not in this case.

Essential to daily operations

My team uses this instrument on a daily basis. We predominantly do upstream bio-process development for our own products as well as customer projects on viral vaccines, AAV, MSCS and iPSCs. Spinner flasks and stirred tank bioreactors provide a significant challenges in establishing a process, with a cornerstone in the cell biology and cell counting. The NC202 reduces the subjectivity experienced with other bench-top cell counting methods and instruments. After initial cell-specific protocol development, internal and external technology transfer has one less barrier. Every biotech and biopharma we work with, uses a NucleoCounter. Ease of use and training on this technology is another bonus. Biotechnology should take a lesson from the justice system: "Eye-witness testimony is the most flawed, but catching the events on a CCTV..." The above strongly offsets the cost and waste generation from the cartridges if there are high sample numbers to analyze.
