Analyses of antibody drugs using ultra high performance liquid chromatography

Antibody drugs, such as monoclonal antibodies (mAbs) and antibody-drug conjugates (ADCs), are known to offer high therapeutic efficacy and reduced side effects due to their specificity and affinity for target molecules. However, because these biopharmaceuticals are manufactured using animal cells, ensuring their structural homogeneity can be challenging. Discover how the ultra high performance liquid chromatography (UHPLC) system, Nexera XS inert, compares to other chromatography techniques including size-exclusion chromatography (SEC), ion exchange chromatography (IEX), and hydrophobic interaction chromatography (HIC), for quality control in the analyses of mAbs and ADCs.

Application NoteSeparations

Efficient method development of monoclonal antibody size variants by SEC

ICH Guideline Q6B requires the separation of impurities such as aggregates and fragments in antibody drugs and determines their content. Therefore, monitoring these impurities by size-exclusion chromatography (SEC) is one of the most important analyses during the production of monoclonal antibodies (mAbs).

Explore this example of efficient method optimization for providing enough separation and robustness for mAb size variants by using the corrosion-resistant Nexera™ XS inert system, pH monitor (pHM-40) and Shimadzu's LabSolutions™ MD software. The method achieves reliable separation while minimizing system damage from high-salt mobile phases.

Optimizing oligonucleotide analysis with UHPLC

Oligonucleotide therapeutics are small synthetic nucleic acids that can be used to treat different types of diseases. In this infographic, curated by Shimadzu, explore the tools available for oligonucleotide analysis, enabling reliable and accurate quantitation to support the development of personalized therapeutics for previously 'undruggable' targets.

Culture medium analysis for a metabolic analysis of antibody-producing cells using LC-MS/MS and ICP-MS

This application note from Shimadzu describes how to use LC-MS/MS (LCMS-8060NX) to analyze 144 organic compounds and ICP-MS (ICPMS-2030) to analyze 9 inorganic elements in the culture medium and culture supernatant of a CHL-YN antibody producing cell line culture. This app note also describes how to use this data to identify which components and metabolic pathways are involved in antibody production.
