It is very clear why so many laboratories are kitted out with Waters LC products: It is reliable, gives reproducible results, and processing data is a breeze which is important in a fast pace environment. We recently underwent an enormous international proficiency scheme and did exceptionally well. This can be attributed to the accuracy of the MS and stable LC quaternary pump system. After-sales is a problem for us as we are based in Sub Saharan Africa - we are therefore limited by Waters to make use of South African agents which has not always given us the standard which we would expect from Waters. The value for money cannot be over-emphasized. Yes, the instrument comes with quite a price tag, but with such reliability and stability, this is worth every cent.

Several challenges exist with quantification analysis of cocaine in post mortem samples in the field of forensic toxicology. Rapid half-life due to hydrolysis within hours of death is just one of these. While difficult to quantify in blood, urine and soft tissues, alternative matrices such as hair and bone can be useful in detecting chronic drug use in post-mortem toxicology cases. A robust extraction and cleanup method is required to efficiently extract drugs from complex matrices like bone, reach a target limit of detection and maintain instrument performance. The use of UPLC-MS/MS has allowed analysts to detect trace levels of analytes by following this method.
