TetraSpeck™ Microspheres, 0.1 µm, fluorescent blue/green/orange/dark red

Thermo Fisher ScientificT-7279Available: Worldwide

The 0.1 µm TetraSpeck™ microspheres are stained throughout with four different fluorescent dyes, yielding beads that each display four well-separated excitation/emission peaks - 360/430 nm (blue), 505/515 nm (green), 560/580 nm (orange) and 660/680 nm (dark red). These microspheres can greatly facilitate the calibration of conventional fluorescence microscopes, confocal laser scanning microscopes, and associated image-proce…

Application Area:

Testing microscope systems, providing fiducials for image registration

These beads are a must for correcting chromatic aberration and sample drift in high-resolution microscopy. The 0.1 µm size is good for measuring point spread functions. They're easy to use -- dilute, spread on a coverslip (or on cells), and mount. Usage tips: Vortex or sonicate to resuspend, and test several dilutions. Beads stick to the coverslip best when dried down, but they can be deformed when over dried. Add mounting media and coverslip them as soon as they are dry. The up-front cost is rather high but I typically dilute 1/1000 or more, so each tube lasts a long time. The fluorescence is fairly stable, so a well-sealed slide is good for several weeks. I did not notice any variation between lots. Tech support for the Molecular Probes family of products is excellent.

These microspheres can greatly facilitate the calibration of conventional fluorescence microscopes, confocal laser scanning microscopes, and associated image-processing equipment for both scientific and commercial imaging, especially for multicolor applications.
