A simple and versatile equipment for quantifying proteins, peptides, and nucleic acids in a high throughput format. Customer service is excellent and they are ready to help with the purchase of consumables and other related accessories. I consider lunatics to be an instrument purchase in money- well-spent category.

Analyze protein and DNA concentrations

We purchased two units, one is the DropSense 96, and the other one is newer model of Lunatic. The DropSense has been working very well for measuring the DNA/RNA and protein concentrations, a full 96-well plate can be read in less than 5 minutes. The results are very reproducible and it’s easy to use. So, we decided to get a newer model and integrate into the Tecan Automation. The software support after-sale was outstanding, the Software engineer took time to walk us through the whole process which enabled us to set up the Tecan Driver Frame work rapidly.

Peptide concentration determination

The device is exceptionally useful for peptide concentration determination prior to LC-MS/MS analyses. It allows us to quickly (5 - 10 min) and without too much sample loss (2 µL) calculate the volume for each individual sample to ultimately inject the same amount of peptide material between a batch of samples. This has a profound effect on the quality of our spectra and reduces the number of re-runs.

DNA quantification

We use the little Lunatic daily to quantify DNA samples (PCR products, Plasmids, genomic DNA): about 200 measurements a month. It is very easy to handle. We very rarely need to re-do a point. It is such a gain of time (Quantifying DNA time was cut 80% roughly!). In 12 months we had one minor breakdown, that was fixed within a week and very efficiently managed by the after sales team. Only downside: I'd love to see recycled plastic chips to improve the CO2 footprint (if it was technically possible). Nonetheless, we're so happy about it that we are buying a second one for our new facility.

Nucleic Acid quantification, Plasma QC

Currently using the Little Lunatic rather than the full scale. I really like the ease of use and the fact that it uses slides to avoid cross contamination. However, I do not like that you have to reload samples to run different applications for analysis. It is a waste of sample and I feel that the machine should be able to reanalyze samples. The Plasma QC application is a game changer! It is a definitive way to determine if a sample is hemolyzed, etc.

Off to the Races: Rapid Buffer Exchange, Protein Quantification and Stability Screening with Junior, Lunatic and Uncle

The conformational, chemical and colloidal stability of a protein are strongly influenced by the buffer solution. Altering buffer salts, pH, ionic strength, excipients and surfactants may increase or decrease the stability of a molecule. The process of screening formulation buffers to optimize stability is a time consuming process. The time required to perform buffer exchange is often a limiting factor in biologics development. Junior with buffer exchange removes the buffer exchange bottleneck by automating buffer exchange and sample concentration, which reduces required hands-on time and increases the number of samples that can be prepared in a day.

Quantification of Protein Samples Using Colorimetric Assays

This application note describes how to use the colorimetric assay applications on the Lunatic systems. Protein quantification is often done by measuring absorbance at 280 nm, results however might be influenced by buffer or source constituents. An alternative to A280 quantification are colorimetric protein assays. Reagent(s) and proteins are mixed, producing a color change which is a measure for the amount of protein present. Unknown sample concentrations are calculated using a measured standard curve.

Quantification of Mammalian gDNA

This application note describes how to use the DNA mammalian application on the Lunatic systems. This Unmix application is used to analyze the UV/ Vis spectral shape of the sample to isolate the fraction of the molecule of interest from co-absorbing entities contributing to the total UV/Vis absorption spectrum. Quantification of Total Nucleic Acids from Unknown Sources (RNA eq)

This application note describes how to use the Total Nucleic Acids application on the Lunatic systems. This Unmix application is used to analyze the UV/Vis spectral shape of the sample to isolate the fraction of the molecule of interest from co-absorbing entities contributing to the total UV/Vis absorption spectrum. Accurate quantification of the molecule of interest is established using its isolated spectrum fraction. This application does not distinguish RNA from DNA, but rather isolates the total nucleic acid concentration in RNA extractions from unknown sources (bacterial, yeast, plant, plasmid, etc.).
