Unlike traditional preparative LC column packing methods, the Axia packing method offers increased sorbent bed density for increased performance and eliminates media bed collapse as a source of premature column failure in preparative HPLC/SFC columns.

Improving Preparative HPLC Column Lifetime in High-Throughput Open-Access Purification Environments

When subjected to viscous samples under high-speed chromatographic conditions, the media bed of the column may deteriorate. This application note describes the use of Axia column technology to eliminate column voiding or chanelling, in conjunction with the SecurityGuard PREP system that protects the column from contaminants and particulates. This work demonstrates the dramatic increase in column lifetime achievable with these systems in place, even when operating under high flow rate, high-pressure conditions.

Increase Column Performance and Lifetime in Peptide and Protein Purification Using Aggressive Wash Conditions on the Ultra-Stable Gemini® Sorbents

Gemini sorbents were specifically developed for chromatographic conditions that demand a wide pH range (pH 1-12). This application demonstrates how the high pH stability of the Gemini product line can be used to extend column lifetime by column regeneration using aggressive alkaline washes (1 M sodium hydroxide in alcohol 1:1 (V/V)). Column regeneration using caustic washes is especially useful in the case of peptide and protein applications where aggregation and impurities tend to accumulate and generate high backpressures. Combining the high pH stability of Gemini particles with the award winning Axia™ pre-packed preparative column will provide longer lifetime and higher performance for the preparative purification of peptide and proteins.

Scaling from Analytical to Preparative Chiral Chromatography While Balancing Purity, Yield, and Throughput under HPLC and SFC Conditions

The work presented in this application will address the relationship between both normal phase and SFC chiral methodologies at the analytical and preparative scale. The impact on resolution at both scales due to flow rates will be evaluated and compared between SFC and normal phase. The effect of preparative column hardware technology along with resulting purity and throughput from related SFC and normal phase purification methodologies will also be evaluated. Axia™ preparative column technology along with Axia specialized hardware shows higher performance than traditionally packed standard hardware preparative columns. The Axia packing technology is compatible with both SFC and HPLC conditions. In this application, we will demonstrate how the Axia packed columns with the Lux® Cellulose-1 polysaccharide-based chiral stationary phase can be a tool to increase throughput for purification of chiral compounds.
