nanoDSF is an advanced dye-free Differential Scanning Fluorimetry method for measuring ultra-high resolution protein stability. By analyzing changes in the fluorescence of the amino acids tryptophan or tyrosine, chemical and thermal stability is assessed in a truly label-free approach. The dual-UV technology by NanoTemper allows on-the-fly fluorescence detection, providing unmatched scanning speed and data point density. This results in ultra-high resolution of unfolding curves, which are not compromised by aggregation thus allowing detection of multiple, and even minute unfolding signals.

 The Prometheus NT.Plex can simultaneously detect protein aggregation with its unique backreflection optics. Using our new NT.Robotic Autosampler, the Prometheus NT.Plex can be fully automated. This setup allows for unattended sample preparation, handling and measurement, and enables walk-away operation with a virtually unlimited number of samples. The modular design of this automated nanoDSF system provides unparalleled flexibly and can thus be adapted to the users’ requirements regarding its technical configurations. For example, the autosampler does not only support automated sampling for up to three Prometheus NT.Plex instruments, but can also be used with our Monolith NT.Automated, or combinations of both instruments. Distinct software packages support straight-forward analysis of thermal and chemical unfolding data as well as develop ability studies.
