In this application note, Biotage describes the use of the SPE-DEX® 5000, a system for automation of disk solid phase extraction (SPE). Three different disk media types are evaluated to better understand the performance of each for a wide range of polycyclic aromatic hydrocarbons (PAH) compounds.
Drinking Water Extraction using High-Capacity SPE Disks
For more than 25 years now, the US has been testing for organic compounds in drinking water using US EPA Method 525. The most common version of this method, Method 525, is a diverse method with more than 100 compounds listed in a wide range of chemical groups such as pesticides, polychlorinated biphenyls, and polyaromatic hydrocarbons. Method 525 offers other changes to the method that may also be of use in making the method more rugged. Additional compounds are evaluated and may be added to the method as the regulatory list is amended every few years.
Automated Solid Phase Extraction (SPE) and GC/MS Analysis of Pond Water Samples According to EN16691 for PAHs
This application note demonstrates the performance of automated SPE for the extraction of PAHs from surface water sampled from a nearby pond containing some particulate. An automated system with DVB disks, following the requirements of method EN 16691 was used.

Triphenyl Phosphate Measurement in Drinking Water using Automated Solid Phase Extraction
Triphenyl phosphate is used as a plasticizer and flame retardant. It is found in foam and televisions as a flame retardant, in candy wrappers, nail polish and other consumer products. It has come into more common use as a safe alternative to polydibrominated ether (PDBE) compounds which were good flame retardants, but found to be a health hazard and have been phased out in many materials. Triphenyl phosphate has become a chemical of concern in several countries. In the US there are airborne exposure limits developed by OSHA. Although triphenyl phosphate is not on the California prop list, which would require listing in public places, there is concern that human exposure may be significant. Recent studies have also indicated concern about possible endocrine disruptor capability. A recent study found that triphenyl phosphate can be absorbed into the body from nail polish. The study found that levels of a metabolite, diphenyl phosphate were elevated seven fold in urine several hours after the nail polish was applied.
Validation of Horizon Technology Disk Extraction Technology for US EPA Wastewater Method 625.1
This application note presents the data collected as part of the demonstration of disk solid phase extraction validation for US EPA method 625.1. Nine different wastewater matrices were evaluated and tested against the criteria listed in Table 6 of method 625.1. The analytes chosen for evaluation were from Tables 1 and 2 in method 625.1, with a few extra analytes from Table 3. The analytes from Table 3 are omitted in this note because there was no acceptance criteria for comparison.
What is Solid Phase Extraction (SPE)?
Solid phase extraction (SPE) is a method of sample preparation that separates and concentrates analytes from solution prior to analysis. In this video find out more about how SPE works, comparing SPE to liquid liquid extraction (LLE).
