Versatile system
Assay and Related Substances on small molecules
The system has 2 flow paths and allows you to perform "normal" HPLC as well as entering the space of UHPLC. With the increase in pressure limits, using columns with smaller particle size is much easier. Although the system is not a "true" UHPLC, it allows one to see the impact of system dwell volume and as a result understand the impact on your separation.

Our lab currently has three ACQUITY Arc systems, with plans to add more in the immediate future to increase capacity. The systems run constantly and are critical to the business. These systems are highly effective in meeting the analysis needs of our lab, and I really don't think we could function without them. Compared to competitor HPLC systems that these replaced, the quality and integrity of the data is much higher, reproducibility was also improved. Analysts find the ACQUITY Arc system easier to use and less time is required to train users to a level of independence using the systems. For these reasons, ease of use received a 5-star rating. Post-sale service and support from the local Waters' service team is legendary! Service is prompt, thorough, courteous and compliant. Working in a regulated environment, the support after the sale is paramount and Waters' delivers exceptional support! For these reasons, both value for money, and after-sales care receive a 5-star rating. Looking at the complete package - instrument, software, support, qualification - Waters' offerings are, in my honest opinion, a fantastic value and worth every penny!

The ACQUITY Arc System is a quaternary-based, modern LC system for scientists working with established methods who are looking for the versatility and robustness required to bridge the gap between HPLC and UPLC while continuing to support validated assays.
The ACQUITY Arc System can replicate your established HPLC methods by compensating for these differences with unique features such as the Arc Multi-Flow Path technology and Gradient SmartStart. The ACQUITY Arc System is enabled by Arc Multi-flow path technology, novel technology that enables the user to easily switch between HPLC and UHPLC operation by selecting the dwell volume that best fits the column separation. System reliability and robustness ensures product consistency and compliance with regulatory guidelines, all while minimizing downtown and inefficiencies.
With the ACQUITY Arc System, maximize productivity through combining system dispersion characteristics with efficient and rapid 2.x um UHPLC separations. Or, accommodate larger, 3.0 - 5.0 um particles. This provides the flexibility to support a wide-range of LC applications.

- Quaternary solvent blending. Blend up to four solvents in any combination or proportion. Expand to up to nine solvent choices with the optional and integrated solvent-select valve for even greater flexibility.
- Plug-and-play method compatibility with HPLC or UHPLC with the flip of a switch. Easily replicate or improve established methods by simply selecting Path 1 (HPLC) or Path 2 (UHPLC) without any manual user intervention. Automatically counteract differences in system dwell volume without the need to alter gradient table inputs.
- Minimize carryover (<0.002%) by continuously cleansing the needle during the run with an advanced flow-through-needle design.
- Flexible column management, providing stable, uniform temperature management with integrated pre-heating options to ensure method repeatability from lab to lab. Optional and integrated column selection valve provide fast, unattended column changeover.
- Directly program gradients in units of pH and ionic strength with Auto●Blend Plus Technology, providing the ability to vary composition in even the smallest increments of pH. This eliminates the cumbersome and error-prone approach of manually preparing mobile phase solutions.

Fast Analysis of Isoflavones in Dietary Supplements – Method Transfer to an ACQUITY UPLC H-Class System
In this application note, the efficacy of different methods for the determination of isoflavones in dietary supplements was determined. The standard methods are based on reversed-phase high-performance liquid chromatography (RP-HPLC) and ultraviolet and visible light (UV-VIs) spectroscopy detection. By transferring a new isoflavones standard method onto the ACQUITY Arc UHPLC System, the run time of the analysis was cut from 70 minutes to 18 minutes.

Demonstration of USP Quetiapine Fumarate Impurities Method Across a Wide Range of LC Systems
In this application the USP quetiapine fumarate impurity method is analyzed on three different LC systems including the Alliance HPLC System, the ACQUITY Arc UHPLC System, and the ACQUITY UPLC H-Class PLUS System. The successful execution of the method on each system will be evaluated based upon the system suitability requirements listed in the USP monograph.
Transfer of USP Assay for Quetiapine Fumarate Across Different LC Systems
In this study, the USP assay method for quetiapine fumarate, an anti-psychotic drug, will be evaluated on a range of systems including an Alliance HPLC System, an ACQUITY Arc UHPLC System and an ACQUITY UPLC H-Class PLUS System. The USP method for quetiapine fumarate was successfully transferred across three systems: an Alliance HPLC System, an ACQUITY Arc UHPLC System, and also an ACQUITY UPLC H-Class PLUS System. All three instruments were able to reproduce the percent of quetiapine fumarate in an unknown sample and meet system suitability requirements, including resolution, tailing factor, and repeatability specifications required by the USP quetiapine fumarate assay method.
Scaling of a USP Assay for Quetiapine Fumarate Across Different LC Systems
In this study, the Waters Columns Calculator will be used to scale the USP monograph assay method for quetiapine fumarate an anti-psychotic drug, to smaller particle sized columns.
Utilizing the method scaling guidelines within the USP General Chapter <621>, traditional isocratic HPLC methods are capable of being scaled to columns with smaller particle sizes and shorter lengths. Scaling a USP method enables the use of modern chemistries and LC hardware to deliver improved throughput with decreased solvent consumption all while providing accurate and reproducible chromatographic data. The scaled modifications for the USP quetiapine fumarate assay method enabled the original method run time to be reduced by 57% for a 3 μm column on an ACQUITY Arc UHPLC System and 87% reduction for the 1.7 μm column on an ACQUITY UPLC H-Class PLUS System. The scaled methods maintained the same or improved chromatographic performance in terms of resolution, peak tailing, and retention time and peak area RSD.
Determination of Isoflavones in Dietary Supplements: A Comparison of Mass Detection with UV Detection
This study demonstrates that in the analysis of complex samples, such as plant extracts, the use of mass detection greatly reduces the interferences from co-eluting compounds. Also, time-consuming troubleshooting for out of specification results are reduced, which will in turn improve overall lab productivity and analytical data quality.
