A Unique Approach to DNA Extraction and Assay Miniaturization
21 Feb 2019
In molecular biology, DNA purity and yield hugely influence the quality of results from downstream applications. In this video, Ben Meadows of LGC, Biosearch Technologies, discusses how the oKtopure® facilitates high-throughput DNA extraction from heterogenous samples. He also highlights how the company’s unique ArrayTape consumable enables rapid, fully-barcoded, assay miniaturization - down to 0.8 µl - in downstream analysis platforms such as the IntelliQube® RT-qPCR system.

Array Tape™
LGC Biosearch TechnologiesArray Tape is a thin (0.3 mm) and flexible continuous polymer strip, serially embossed with reaction wells in customised volumes and formats including 96- and 384-well array. A reel with 40 meters of Array Tape carries 277 equivalents of a 384-well plate (106, 368 reaction wells). Array Tape is the ideal consumable for fast, streamlined and continuous lab workflow processes.

IntelliQube™ integrated PCR workflow
LGC Biosearch TechnologiesFully Automated PCR Setup, Amplification and Analysis System. The IntelliQube is the first fully automated and high-throughput instrument featuring seamless integration of liquid handling, thermal cycling, detection, and data analysis. The IntelliQube supports quantitative real-time PCR, end-point PCR, and isothermal chemistries, allowing you to realize the benefits of inline automation enabled by the innovative Array Tape™ consumable. IntelliQube leverages best-in-class multi-functional dispensing for high speed liquid handling in 384- or 768-well Array Tape™. Precise sample dispensing into Array Tape is handled by an interchangeable CyBi® FeliX 96- or 384-channel pipetting head. The IntelliQube supports inline amplification with a Peltier thermal cycling block designed specifically for Array Tape™. Due to a unique block design and surface geometry, both 384- and 768-well Array Tape are supported, without requiring a block change. The IntelliQube features an advanced optical system optimized for inline fluorescence detection in Array Tape. Filtered LEDs are optimized for a variety of commonly used fluorophores excited within the range of 480-620 nm. A high-resolution CCD camera supports detection of fluorescence intensity within 5 detection channels for dyes emitting within the range of 510-705 nm. Multiplex data capture for all 5 optical channels can occur in as little as 10 seconds. Benefits: Walk-away operation Increased efficiency of lab personnel Reduced chemistry costs Increased throughput Reduced consumable costs

oKtopure™ - Automated nucleic extraction platform
LGC Biosearch TechnologiesThe oKtopure™ is a fully automated nucleic acid extraction instrument that enables a new level of high-throughput processing and significant cost savings. This platform combines standardised high-throughput DNA or RNA extraction from a variety of sample types with the proprietary sbeadex™ magentic bead based extraction chemistry. The oKtopure is part of our on-going programme to provide customers with high productivity solutions, first developed and optimised for use within our service labs, where every day we deliver Extraction, Sequencing and Genotyping services. Key to the delivery of consistently high quality and high yield DNA preparations from the oKtopure is its ability to bind and hold the magnetic beads, and hence the DNA in place whilst transferring the buffers required for washing, binding and elution steps. The final aqueous wash used in the sbeadex extraction protocols enables the delivery of nucleic preparations ideal for downstream applications such as SNP genotyping, NGS and Sanger sequencing. Features: High throughput - capable of processing eight 96-well plates (768 samples) in approximately one-and-a-half hours Robust tip washing system reduces consumable costs by 50% or more Preloaded methods for different sample types – fully customizable Extraction expertise and support from scientists who use the instrument daily and have been carrying out high throughput extractions since 1996!
