Application Area:

High & low molecular weight heparin

The columns are easy to use but take care when doing so. Loss of resolution on the low molecular weight region (column bench 3000 + 2000) occurs very quickly, especially in mobile phase sodium phosphate. Ammonium acetate buffered at pH 6 so they are more stable. It seems like some particular stationary phase batches are more stable and reproducible than others. These columns have never been cheap.

TSKgel SW-type columns for size exclusion/gel filtration analysis of antibodies and other proteins are silica-based. A hydrophilic diol-type bonded phase shields the silica surface from interacting with protein samples.

TSKgel SW-type columns contain more pore volume per unit column volume, which results in higher MW selectivity or better resolution. In addition, they show low adsorption and well-defined pore size distribution, which are requirements for high performance GFC of proteins.

Various pore sizes are available in TSKgel SW, SWXL, SuperSW, UltraSW and UP-SW product lines.

Size Exclusion Chromatography

Size Exclusion (SEC) is the general name for the chromotographic mode also referred to as gel permeation chromatography (GPC) for non-aqueous elution systems or gel filtration chromatography (GFC) for aqueous systems.
SEC is a method in which components of a mixture are separated according to their molecular size. Large biomolecules that cannot penetrate the pores of the packing material elute first and flow with the mobile phase in the interparticle space of the packed column.
Smaller molecules can partially or completely enter the stationary phase and will elute from the column after the excluded sample components.
SEC is a very simple method for separating biomolecules, because it is not necessary to change the composition of the mobile phase during elution. However, the separation capacity of this method is limited.
