"The kit is relatively easy to use for a person who knows how to pipette small quantities of solution accurately. As we used it with students, one kit was just spent on getting the pipetting right, as well as determining the dilution factor of the sugar cane juice as the kit can only measure amounts in the nanomole range (0.2-1 nmol for the fluorometric assay and 2-10 nmol for the colorimetric assay). At a price of more than AUD$600, this is not cheap. Once we had the pipetting and dilution under control, we used another kit, each of which is said to do 100 tests. However, because you always have to do a calibration curve on each plate, you lose some capacity. We always did at least five calibration points in case one or the other don't work (a note besides, you do not need to do the free glucose test for the calibration curve, as the standards contain none) Overall the analytical process is manageable even for students, however, we had severe problems using the Technical Instruction Bulletin for the calculation which is necessary at the end of the analysis. Therefore, I only give the kit four points. When contacting the company about that we got a swift response, but still struggled a bit with the interpretation of the results. I think this kit is very well suited for Glucose/ Free Glucose and Sucrose analysis, and the price is comparable to other suppliers, however, the instructions should be improved."

Sucrose is a disaccharide that is hydrolyzed to glucose and fructose by invertase. Glucose is the primary carbohydrate utilized for energy during cellular respiration. Upon cellular uptake, glucose can be shunted towards breakdown via glycolysis or polymerized for energy storage.

In this assay kit, glucose is oxidized via glucose oxidase resulting in a colorimetric (570 nm)/ fluorometric (lex = 535 nm/lem = 587 nm) product, proportional to the glucose present. To measure sucrose, invertase is added to the reaction to convert the sucrose to glucose and fructose. The free glucose can be subtracted from the total glucose to give the concentration of sucrose present. This kit has a linear range of detection between 0.2–1.0 nmole of glucose for the fluorometric assay and 2–10 nmoles of glucose for the colorimetric assay.

This kit is suitable for use with various biological samples including serum, plasma, other body fluids, food, and growth media.

  • 96 well flat-bottom plate – It is recommended to use black plates with clear bottoms for fluorescence assays and clear plates for colorimetric assays.
  •  Fluorescence or spectrophotometric multiwell plate reader.
