SamuxMP
The Refeyn SamuxMP is a mass photometer optimised for adeno-associated virus (AAV) characterisation and is an essential analytical tool for laboratories working with AAVs. The SamuxMP precisely measures the empty/full capsid ratio for AAVs of any serotype. SamuxMP mass photometry measurements are rapid and require very little sample.

The Refeyn SamuxMP is a mass photometer that is tailor-made for the needs of AAV development and quality control. It can clearly resolve the difference between empty and loaded AAV capsids, providing a precise measurement of the empty/full capsid ratio of an AAV sample in just minutes with very little sample. It can also reveal the presence of unwanted species, such as partially filled capsids, overfilled capsids or aggregates.
The SamuxMP fits on a laboratory benchtop and can be operated easily by a user with basic lab skills. It requires very little sample and works fast. A single measurement – including data analysis – can be completed in under five minutes. Designed for easy decontamination and to prevent liquids from entering the machine, the SamuxMP is well suited for work with AAVs.

Optimizing and monitoring AAV capsid purification using mass photometry
This application note details Refeyn's collaboration with Pharmaron Gene Therapy to demonstrate the capabilities of the SamuxMP mass photometer in adeno-associated virus (AAV) analysis. The SamuxMP is optimized for AAV analysis and proves instrumental in process development and downstream purification monitoring. It quantifies the proportions of empty and full capsids and detects impurities. Using mass photometry technology, the SamuxMP accurately measures individual particle masses regardless of size or shape, providing quick results with minimal sample requirements. Pharmaron Gene Therapy utilized the SamuxMP to analyze AAV samples at various development stages, from harvest to chromatographic column separation.
Application of mass photometry in GMP-regulated environments for characterization of AAV samples
In this whitepaper, Refeyn defines how the SamuxMP and SamuxMP Auto can be applied in a GMP-regulated environment and outlines the key features of the SamuxMP software package. The whitepaper addresses challenges in the commercial manufacturing of adeno-associated viruses (AAVs), commonly used vectors for gene delivery. It highlights the limitations of current AAV characterization methods, such as analytical ultracentrifugation (AUC), electron microscopy (EM), or qPCR/ELISA. Mass photometry is introduced as a solution that overcomes bottlenecks, providing reliable data at the single-particle level and aligning with traditional characterization methods.
AAV analytics with mass photometry
Mass photometry is an innovative technology that analyzes biomolecules, including adeno-associated viruses (AAVs), at the single-particle level, in their native state and without the need for labels. In this application note, Refeyn shows that mass photometry is an ideal analytical technology for AAV research, development and manufacturing. In just minutes, a mass photometer can precisely measure the empty/full capsid ratio for AAVs of any serotype. It requires only very small sample volumes and concentrations, and minimal sample preparation.
Mass photometry of AAVs
In this application note, Refeyn shows how mass photometry can be used across AAV serotypes to reliably and reproducibly measure AAV sample purity and cargo loading. These measurements can be performed in minutes with small quantities of sample.
