The TempliAMP™ RCA Kit is a versatile Rolling Circle Amplification (RCA) kit that is used with a circular DNA template and short DNA primers to continuously synthesisze DNA around the circular template. Amplification of DNA is through an isothermal reaction accomplished using using Phi29 DNA polymerase, a high fidelity, high-processivity replication enzyme, thus, thermal cycling as in PCR in not required. The TempliAMP™ RCA Kit can be applied to various types of circular DNA templates, such as (1) amplification of purified circular DNA and (2) direct amplification of circular DNA from bacterial sources, such as plasmids. For bacterial sources, this protocol eliminates the need for DNA extraction, allowing RCA of circular DNA templates directly from colonies on agar plates, glycerol stocks and liquid medium cultures.
