Natural products screening
We use the cytometer for general purposes and, specifically, to run high throughput screening assays for antibiotic and antifungic activity of natural products. It can run plates of 12 up to 384 wells, and even you can define 12-48 eppendorf tubes acquisition setup, meaning that it's very versatile. The speed of run depends on the target, the cell type, the number of events to acquire and the viscosity of the natural product to test (the washing steps between wells, increase). In normal conditions 96 well plate, acquiring 10000 events per well takes 35 minutes in our hands, with less than 0.11 of carryover. It allows us to run libraries of hundreds of compounds per day. The system is robust, and the technical service is very good, keeping the instrument in its best shape. As analysis software we use Flowlogic, that provide fast plate analysis and easy lead detection. MACSQuant X worth its price.

- Measurement speed: < 15 minutes per 96-well plate (5 µL measurement volume per well) Referred value indicates the average of multiple experiments and can differ for individual sample materials. The measurement speed is determined by measuring the time between the movement of the robotic arm into the first measured well, and its movement out of the last measured well. The measurements were carried out at high flow rate in fast mode

- Sample carry-over: 0.1 % Referred value indicates the average of multiple experiments and can differ for individual sample materials. For carry-over, full 96-well plates were loaded with 200 μL/well of PBMC suspension at a nominal concentration of 5,000 cells/μL in every other well ("SRC-wells”). Alternating wells are loaded with an equal volume of MACSQuant® Running Buffer ("CO-wells”). The uptake volume was set to 50 μL at medium flowrate. Plate shaking was set to 1200 rpm and a singlet gate was used to determine singlet counts in originating wells as well as in carry-over wells. The carry-over is defined by sum (CO-singletcount)/sum(SRC-singletcount) ×100%. Results for 384-well plates do not differ significantly from those measured in 96-well plates.

MACSQuant X Analyzer Facilitates Ca²+ Flux Assays
Intracellular calcium (Ca²+ ) serves as a crucial second messenger regulating a variety of physiological processes such as cell division, muscle function, neurotransmission, and vascular contraction. Upon specific stimulation, intracellular Ca²+ concentrations can quickly and significantly increase by the release of internal Ca²+ storages or by entry from the extracellular space through ion channels. Measuring this Ca²+
