We have been using cleanNGS for several years now at ILVO for clean-up of Illumina targeted sequencing libraries and clean-up of Nanopore WGS libraries. It replaces the AMPureXP beads 1:1 for several workflows and we are very happy with the consistently high quality, good value for money and simple protocol.

Purification PCR material for Miseq
Purification of DNA with clean NGS is time efficiënt compared to the use of purification columns. The protocol is straightforward and easy to follow. You can purify more samples at once, so it is handy for high throughput. The total cost gets lower the more samples you process.

Extraction and cleanup of plant DNA
At our lab the magnetic bead based plant DNA purification system Clean Plant PK DNA Kit is routinely used for quick and robust gDNA isolation from various plant species. The ease of use and compatibility to our liquid handler made it one of our most preferred plant DNA extraction protocols. After implementing the CleanNGS magnetic beads in our lab routines we covered most of our plant DNA handling and purification methodologies. The CleanNGS magnetic beads substitute many of other manufacturers magnetic beads with comparable or even better results when applied for size selection and purification of our plant DNA extracts. Clean NGS products helped our lab to improve quality and throughput and substantially cut costs and hands-on-time.

Clean up libraries for next generation sequencing
Some members of the lab were not keen to change away from the regular bead clean up supplier. We tested these and found they gave slightly higher amount of DNA and just as clean.

The CleanNGS kit offers a highly efficient magnetic bead based clean-up system for the purification of both DNA and/or RNA for next-generation sequencing workflows. CleanNGS provides maximum flexibility allowing for left, right or double-sided size selection by easily adjusting the sample to CleanNGS volume ratio(s).
Based on our proprietary chemistry CleanNGS removes, salts, primers, primer-dimers and dNTPs, while DNA and/or RNA fragments are selectively bound to the magnetic particles based on their size. Purified DNA and RNA is eluted of the magnetic particles using water or a low salt buffer and can be used directly for downstream applications. The protocol can be adapted to your current liquid handling workstation (e.g. Hamilton, Beckman, Agilent, Caliper, Perkin Elmer, Tecan and Eppendorf) utilizing your current protocol as well as being used manually.

Performance of CleanNGS in next-generation sequencing
Next generation sequencing (NGS) allows for the generation of robust information on the genetic diversity of organisms at the single cell, organ, species or population level. To perform an NGS experiment, users must prepare a sequencing library from a purified nucleic acid sample. Magnetic beads have many uses within NGS workflows. In addition to nucleic acid isolation and purification, magnetic beads are also used for size selection in NGS library preparation and library normalization. With emerging NGS technologies, there is an increased need for NGS library purification methods providing accurate results starting from low input amounts of DNA and/or RNA. Since CleanNGS (CleanNA) can be automated, as well as being used manually, it can be adopted in any NGS laboratory independent of sample throughput. In this poster, CleanNA compares the performance of CleanNGS in DNA cleanup versus two equivalent magnetic beads based kits.
