The 1290 Infinity II GPC/SEC System is a complete GPC/SEC analysis solution that is capable of both analytical- or microscale operation. The low-volume refractive index detector enables microscale operation, which offers several key benefits, including shorter run times for higher sample throughput, ultralow dispersion for improved resolution, and lower solvent usage and waste disposal in order to reduce costs.

Coupling with the Openlab GPC software enables archiving of all data together in a single CFR compliant environment.

High-resolution sampling 2D-LC for pharmaceutical impurity analysis

In this application note, Agilent addresses the challenge of analyzing impurities in low concentrations relative to an active pharmaceutical ingredient (API) for quality control purposes. When the impurities have structural similarities to the API and there is a significant concentration difference, achieving chromatographic separation and detection becomes difficult.

To overcome this challenge, Agilent introduces the Agilent 1290 Infinity II 2D-LC Solution, which offers the flexibility to switch between comprehensive (LCxLC), multiple heart-cutting (MHC), or high-resolution sampling 2D-LC (HiRes 2D-LC) techniques. In this specific application note, HiRes 2D-LC is utilized to separate two closely eluting compounds, with one compound present at a very low concentration and concealed beneath the more highly concentrated compound peak. The analysis involves chlorodifluorobenzoic acids as standard substances and deamidated insulin as a representative real sample.

In this application note, Agilent highlights the benefits of utilizing the Agilent 1290 Infinity II micro Refractive Index Detector (RID) for gel permeation chromatography (GPC) within an Agilent 1290 Infinity II LC system. The combination of the ultra-high performance liquid chromatography (UHPLC) system with the micro RID allows for the utilization of GPC columns with narrow inner diameters of 2.1 mm. This setup results in improved resolution and reduced solvent consumption compared to a standard HPLC GPC system equipped with a standard RID.
