Robust Routine Amplification of Long DNA Fragments Using KAPA Biosystems LongRange HotStart and HiFi HotStart

This application note describes two solutions for genetic testing of long amplicons from KAPA Biosystems: LongRange HotStart and HiFi HotStart PCR kits. Some genetic and pharmacogenetic tests require the routine amplification of long DNA fragments from complex genomic targets. Assays targeting fragments >3 kb often require careful optimization of reaction parameters, such as Mg2+, primer and template concentrations and cycling parameters. KAPA LongRange HotStart offers robust amplification in single and multiplex assays, whereas KAPA HiFi HotStart is capable of amplifying human genomic targets >10 kb with an error rate up to 50 times lower than long-range enzyme blends.

The KAPA Long Range PCR system is a blend of Taq DNA polymerase and an engineered archaeal (B-family) DNA polymerase possessing proofreading capability. This two-enzyme system is designed specifically to support long-range and/or sensitive PCR, and possess 3 – 4X higher fidelity than Taq polymerase. In the hot-start formulation, KAPA Long Range is combined with a proprietary antibody that inactivates the enzyme until the first denaturation step, eliminating spurious amplification products and increasing reaction efficiency and sensitivity. KAPA Long Range is also supplied in 2X ReadyMix format with loading dye that contains all the components required for PCR except primers and template. KAPA HiFi DNA Polymerase is a novel, single-enzyme system that exhibits industry-leading performance when compared with other high fidelity polymerases and polymerase blends. KAPA HiFi has been engineered to have an increased affinity for DNA without the need for accessory protein domains. The intrinsic high processivity of KAPA HiFi results in significant improvements to yield, sensitivity, speed, target length, and the ability to amplify difficult templates (e.g AT- and GC-rich).
