High-Resolution Separation of a Fusion Protein

28 Aug 2014

This application note describes a high-resolution hydrophobic interaction chromatography (HIC) method to separate a fusion protein from a truncated version and a structural isomer.

Thermo Scientific™ ProPac™ HIC Protein and Peptide Column

Thermo Fisher Scientific

Hydrophobic Interaction Chromatography Columns for the Separation of Large BiomoleculesHydrophobic Interaction Chromatography separates biomolecules in a decreasing salt gradient, based on differences in surface hydrophobicity. This method preserves the biological activity of proteins. The HIC separation mechanism is complementary to those of ion-exchange and gel filtration chromatography. The ProPac® HIC-10 column is a high-resolution, high-capacity, silica-based HIC column that provides greater hydrolytic stability under the highly aqueous conditions used in HIC. Examples include the separation of bovine serum proteins, snake venom proteins, enzymes, human skeletal muscle protein (HSMP), monoclonal antibodies, pancreatin, and thrombin and peptide applications including tryptic digests of proteins.ProPac® HIC Protein and Peptide Column Features: Based on 5 µm ultrahigh purity spherical silica gel particles, with 300 Å pores Proteins separated under non-denaturing conditions High protein loading capacity for protein purification applications Proprietary multidentate silica bonding for enhanced hydrolytic stability High-resolution HPLC separation of proteins and peptides Wide range of applications Column Details: Phase: HIC Particle Size: 5 µm Pore Size: 300 Å Length: 75, 100, 250 mm ID: 2.1, 4.6, 7.8 mm

Thermo Scientific™ MAbPac™ HIC-10 Monoclonal Antibody Column

Thermo Fisher Scientific

Designed for separation of monoclonal antibodies (MAbs) and related biologics by hydrophobic interaction, the Thermo Scientific™ MAbPac™ HIC-10 LC column is a high resolution silica column. The MabPac HIC-10 column's unique chemistry provides high resolution, excellent biocompatibility, and selectivity that is different from other HIC columns on the market. The MAbPac HIC-10 column is based on high-purity, spherical, wide-pore (1,000Å), 5μm silica particles functionalized with proprietary alkyl amide groups. Its advanced surface bonding technology leads to excellent chemical stability, high efficiency, and unique selectivity optimized for MAbs and different from other HIC phases.Features: Excellent biocompatibility and extremely low carryover High resolution and high efficiency Compatible with both organic solvent and aqueous mobile phases Rugged column stability Column Details: Type: Hydrophobic Interaction, Alkyl amide Particle Size: 5 µm Pore Size: 1000 Å Length: 100, 250 mm ID: 4.6 mm
