Correlation of Mutations Detected in Liquid and Tissue Biopsies Using the Maxwell® RSC

This application note describes a method for purifying nucleic acids using the Maxwell® RSC instrument, based on novel surface and binding chemistries. Circulating cell-free DNA (ccfDNA) in plasma can be used to detect biomarkers that show great promise for diagnosis and monitoring of cancer, giving rise to the possibility of liquid biopsies that obviate the need for invasive tissue collection. This protocol is completely automated, decreasing the chance of user error or contamination and sixteen samples of up to 1ml can be processed in 80 minutes, making next generation sequencing of DNA a viable potential method for screening plasma in a form of liquid biopsy.

QuantiFluor® dsDNA System

The QuantiFluor® dsDNA System contains a fluorescent DNA-binding dye that enables sensitive quantitation of small amounts of double-stranded DNA (dsDNA) in solution.   The quantitation of dsDNA is a very important step in many biological applications, particularly in standard molecular biology techniques. The dye shows minimal binding to single stranded DNA (ssDNA) and RNA, allowing specific quantitation of dsDNA. The QuantiFluor® dsDNA System was developed using the fluorescence module of the GloMax® Multi+ Detection System and the QuantiFluor®-ST Handheld Fluorometer. The QuantiFluor® dsDNA System can be used with any fluorometer that is capable of measuring fluorescence at the appropriate excitation and emission wavelengths, including the Quantus™ Fluorometer. Each QuantiFluor® dsDNA System contains sufficient volume to perform 200 assays at 2ml or 2,000 assays at 200μl. QuantiFluor® dsDNA System Features & Benefits:   Specificity - Highly specific to dsDNA, minimal binding to ssDNA, RNA, protein and interfering compounds. Sensitivity - Significantly increased sensitivity over absorbance at 260nm (NanoDrop® spectrophotometer) for low-concentration samples. Performs better or equal to PicoGreen® dye and can detect as little as 50pg/ml. Ease of Use - System includes all required reagents to quickly set up and quantitate dsDNA. Instrument Compatibility - Pre-optimized on both the Quantus™ Fluorometer and GloMax®-Multi+ Instrument.

Maxwell® CSC DNA FFPE Kit

  The Maxwell® CSC DNA FFPE Kit is used in combination with the Maxwell® CSC Instrument to provide an easy method for efficient, automated purification of genomic DNA (gDNA) from human FFPE breast, lung or colon tissue samples.   The Maxwell® CSC DNA FFPE (formalin fixed, paraffin embedded) kit is intended for use, in combination with the Maxwell® CSC Instrument and the Maxwell® CSC DNA FFPE purification method, as an in vitro diagnostic (IVD) medical device to perform automated isolation of DNA from human breast, lung and colon FFPE tissue samples. The purified DNA is suitable for use in amplification-based in vitro diagnostic assays. Available in the US only.
