SYNTAX System
DNA Script introduces a new paradigm in oligo synthesis, the SYNTAX System, powered by Enzymatic DNA Synthesis (EDS) technology to accelerate life science innovation. Designed for ease of use, productivity, convenience, control, and confidentiality, the SYNTAX System enables automated nucleic acid synthesis within a benchtop solution. In this application note, you can find all applications and benefits of the technology.
SARS-CoV-2 spike protein mutagenesis
Recombinant spike (S) proteins are a critical resource for functional and structural SARSCoV-2 studies. In this application note, DNA Script demonstrates the use of primers produced with DNA Script's novel Enzymatic DNA Synthesis (EDS) technology in a pragmatic, site-directed mutagenesis strategy for the production of optimized and variant SARS-CoV-2 spike protein expression plasmids. EDS primers supported efficient multi-site mutagenesis and Sanger sequencing confirmation of mutations and cloned inserts. Rapid access to custom oligos enabled predictable project schedules amidst a global synthetic DNA supply bottleneck.
SARS-CoV-2 amplicon sequencing
In this application note, DNA Script demonstrates the use of primers produced with its novel Enzymatic DNA Synthesis (EDS) technology for whole-genome amplicon sequencing and phylogenetic analysis of SARS-CoV-2 isolates using the ARTIC network protocol.

Optimization of a single molecule fluorescence in <i>situ</i> hybridization assay (smFISH) for Drosophila A virus detection
The use of same-day enzymatic synthesis to quickly iterate through probe design to fine-tune sensitivity and specificity has been found to accelerate the optimization of smFISH assays and thus acts as a rapid and simple alternative to immunofluorescence. In this webinar, Dr. Steven Quistad, DNA Script, and Bailey Schmidt, DNA Script, will discuss a recent study carried out in collaboration with the Institut Pasteur using on-demand enzymatic DNA synthesis (EDS) to accelerate the design-print-test-learn cycle required to optimize smFISH assays for the detection of viral RNA in the gut of Drosophila melanogaster.
Key learning objectives
- Understand the basics of DNA synthesis (EDS) and the SYNTAX System
- Understand how on-demand EDS probes printed on the SYNTAX System accelerates smFISH assay optimization
- Understand how generating labeled probes with the SYNTAX System accelerates iteration
- Discover the advantages of making on-demand DNA with enzymes
Who should attend?
- Gene expression researchers
- Genomics researchers
