Great replacement for radioactive 2-DG assay.

  • Application Area: Measure glucose uptake in cells

"Great alternative to radioactive 2-DG assays. However, not specific to 2-deoxyglucose and it will detect endogenous glucose in the cells, which could lead to high background. Several additional components are required that aren't obvious until you read the protocol in detail. Normalizing is difficult because of lysis buffer components. Was able to scale down to 384 well format. Functioned in principle, but potentially difficult to use at this scale without good lab practice and tools. Is my first choice for non-radioactive glucose uptake."

Easy-to-use kit, but expensive and not as great results with L6 cells.

"This kit is very straightforward and easy to use. The only down sides are the cost (it is quite expensive) and the fact that for the cells I use, I don't get as strong of an insulin-stimulated glucose uptake response as I'd like (despite the fact that I am following the kit's instructions for L6 cells)."

Glucose metabolism is a key process in many organisms. A lack of insulin-stimulated glucose uptake is associated with type 2 diabetes, while high glucose uptake is a sign of the high glycolytic rates associated with cancer. Measuring glucose uptake can determine the effects of various treatments for diabetes and cancer.

The gold standard method of assaying glucose uptake relies on detection of radio-labelled glucose analogs. Alternative colorimetric and fluorometric glucose uptake assays often lack the sensitivity and robustness needed to reliably measure glucose uptake in cells.
