"The system works well for the purpose of study which is separation of exosome based on size. Although setup and optimization of instrument are quite complicated, the support staff are well-trained and professional. I usually get replies to emails email within 1-3 working days."

AF4 has great potential to fractionationate without the adverse effects of LC/SEC.

"The instrument is designed well for ease of use in batch or being modular for online mode. However, the software needs to be well integrated so that it is a more cohesive software package where all of its capabilities are centralized into one package. As a result the software has a steep learning curve. Also, the AF4 channel design can definitly be improved with torqued latches so that assembly can be done more simplistically by flipping latches instead of manually by hand with a torque wrench."

The Eclipse® AF4 solves the most difficult separation, fractionation and sizing tasks in the macromolecular and colloidal arenas. Because it has no stationary phase, the advantage of asymmetric-flow field-flow fractionation (AF4) over traditional SEC column chromatography is the ability to separate both soluble and colloidal components, no shear and little to no adverse surface interactions. Integrates with industry-leading HPLC modules. 

The Eclipse is most often used with a DAWN® multi-angle light scattering (MALS) detector and ASTRA® software for FFF-MALS-DLS analysis of molar mass, size and conformation distributions.

Features: 

  • Separation range: 1 – 10,000 nm
  • Solvent compatibility: 2 models - aqueous (biocompatible) and organic solvents
  • Supported HPLC modules: The Eclipse AF4 integrates with Agilent, Thermo and Shimadzu pumps and autosamplers as well as degassers, online detectors and mass spectrometers
  • Supported separation channels: 2 sizes of standard-injection analytical AF4; frit-inlet channel for sticky or aggregation-prone samples; Mobility® electrical/AF4 module for zeta-potential analysis; and semi-prep channel for mg-scale separations
  • Software: AF4 and HPLC modules may be controlled from native HPLC software packages with plug-ins, or from the VISION® chromatography software suite which includes SCOUT DPS® for virtual method development and size analysis, and controls ASTRA software for MALS/DLS analysis
