Great liquid dispenser, a must have!

Acoustic compound/reagent dispenser

Echo 550 allows users to dispense compounds or reagents in nL quantity, which not only reduces the amount of compound/regent required for running experiments but also provides a safer way to dispense compounds with unknown safety profiles. The Echo Dose-response software requires some training before use, but the dispense reports generated from the software can be imported into LIMS, which is great for analyzing HTS assays. The Echo Plate Reformat software is very intuitive to use and is a great tool for running any exploratory assay. This software is the most popular amount the users in my lab. The weekly maintenance for this instrument is very minimal, but we do need to reach out to the Labcyte engineer when an error occurs since the instrument is quite complex. Overall, Echo 550 has become an essential instrument for in vitro drug discovery in our lab, especially for running any high throughput screening campaigns.

Cell-based compound profiling

The draw of the Labcyte Echo 555 Liquid Handler is the speed at which compounds can be added to 384-well plates, and the accuracy and flexibility in which it does it for full integration into a robotic system. The ability to add nanoliter volumes in seconds is of incredible value to large batches of plated cells, and the service is impeccable. I cannot imagine trying to screen the number of compounds against multiple cell lines without the Echo.

ECHO is one of my favorite machine in the lab. I can use the machine to dispense small volume ( minimum: 2.5 nl). The machine has 3 different programs: Cherry Pick, Dose response, and Plate reformat. It is so convenient for high throughput screening. It is very important to have ECHO for Drug Discovery because to can test/screen a lot of compounds and use a very small amount of precious compounds. The machine is reliable, save you time, and very efficient. When I show people around the lab, I show them the ECHO first.

The ECHO acoustic liquid handler is easy to use and makes setting up dose-response curves of many samples so much easier and quicker than by hand. It is also much more accurate than using a normal pipette as it dispenses 2.5nl droplets.

Preparation of Mouse Plasma Microsamples for LC-MS/MS Analysis Using the Echo Liquid Handler

Serial microsampling of animals reduces the overall number of animals sampled and euthanized for toxicology studies. Traditional composite studies in mice can require up to 700 μL of blood for each time point in a pharmacokinetic study. A draw of 700 μL from a mouse would require the animal to be euthanized and often require the use of addition “satellite” animals to complete the study. Thus, in a 9-point time course analysis of mouse plasma, nine mice would be sacrificed. In response to programs like the EU 3Rs initiative to reduce the use of animals in research researchers have turned to microsampling as an alternative to composite studies. With advances in bioanalytical techniques drug levels can be determined from samples of 20 μL or less. This small sample volume or microsample enables reduces the dependency on satellite animals to complete a study. Additionally, microsampling is faster and less stressful than traditional composite studies.

Miniaturization of an HTRF Methyltransferase Assay that Detects Histone Modifying Activity

This application note demonstrates the use of an HTRF methyltransferase assay to assess the methyl transferase activity of histone modifying protein SET7/9. The EPIgeneous Methyltransferase Assay kit from Cisbio is used, however the assay is miniaturized to reduce the volume of assay reagents, using Labcyte’s Echo liquid handler and BMG LABTECH’s PHERAstar FS microplate reader. This article shows how miniaturization of assays using precise instrumentation can reduce the cost of reagents but still provide excellent experimental data.

The Development of an Echo® Qualified Reagent Reservoir

To enable the transfer of large quantities of fluid using acoustic liquid handling, Labcyte is developing a companion consumable, the multi-well Reagent Reservoir (RES). This study characterizes fluid transfer from a RES prototype for volumes on the scale of nano- to microliters per destination well. Additionally, to demonstrate the combined performance capabilities of the RES and the Echo® 525, a 2-fold dilution quantitative polymerase chain reaction (qPCR) assay was performed.
