"Cytoflex S allows an easy way to analyze extracellular vesicles (EV) with its impressive 405-SSC and fluorescence sensitivity. Using beads for calibration, it's able to detect 80um beads without problems, but this is not the real thing, as we know. The real thing is to work with biological particles of known size (defined by other technique, DLS, Nanosight or EM) and use them as size standards. We use phages as standard (from 50nm to 150 nm) and Cytoflex S is able to discriminate them from noise easily, therefore, allowing us to analyze EV without problems. It's fluorescence sensitivity lets us detect low epitope EVs. Also is possible to differentiate by scattering of different EVs populations. Its use is very simple, can use 96 well plates, the software is very straightforward and the price, keeping in mind that is a 4 laser/13 fluorescences instrument, is very competitive."

"The CytoFlex has won me over! I was a little sceptical of voltage free detectors, but the users have found this so easy to use, that I realise that I may have been a bit conservative in my own mind. The biggest bonus has been the increased sensitivity which has increased the ease of analysis through being able to discriminate weak positive samples. Another key change for us is the ability to use it for single tubes or for plates. In a multiuser core facility, this has increased the user base significantly and results in long sessions using the plate loader freeing up the user to carry on with other research. Indeed, they often add the plate at the end of the day, go home ad then collect the data in the morning. "We have owned one for just over a year, cost ~$50k for a two laser, 5 color system with plate loader. Very easy to use, quick to set up in just a few minutes and at least as sensitive as our core's FACSAria III. Filters are easily replaceable if you have a specific application but works great out out of the box. I can have members of staff trained on it in just a few minutes. Minimal downtime so far, mainly from poor cleaning as the manual does not have a clear daily startup and shutdown protocol. I would recommend buying a multiple probes and peristaltic tubing as these are the only consumables. We love it, use it daily, and I would buy it again. The software is a hybrid of Diva and FlowJo and produces reasonable quality images so no subsequent analysis required."

Its sensitivity is amazing and it is very versatile, it can be used for microvesicles analysis, microbiology applications, immunophenotyping, plant ploidy and many more applications. Normal configuration is 3 lasers (405, 488, 640 nm) and 13 fluorescence detectors. Also can be equipped with a plate loader (but it needs some improvements). Its price is very affordable for such a high value instrument. It has almost no maintenance. "
