PL-SAX for Biomolecules is ideal for the anion-exchange HPLC separations of proteins and deprotected synthetic oligonucleotides under denaturing conditions. The covalently linked strong anion exchange functionality extends the operating pH range, and the anion exchange capacity is independent of pH. For synthetic oligonucleotides, separations using denaturing conditions of temperature, organic solvent, and high pH are all possible.

PL-SAX for Biomolecules delivers improved chromatography for self-complementary or G-rich sequences that may associate to form aggregates or hairpin structures. The 5 µm material provides high efficiency separations of n and n-1 sequences.

Analysis of oligonucleotides with ion exchange chromatography and Agilent Infinity II Bio LC system

In this application note, Agilent Technologies demonstrates the effectiveness of using its Bio SAX columns for oligonucleotide analysis, which is better suited for certain analyses compared to ion-pair reversed-phase chromatography. The Bio SAX was able to conduct high-resolution ion exchange chromatographic separations of DNA and RNA oligonucleotides over a broad range of sizes without being affected by corrosion or oxidative damage from eluting salts. This application note demonstrates the versatility of ion exchange chromatography for oligonucleotide analysis and purification processes, and it provides a good starting point for further method optimization.
