Multiparametric reporter gene assays
The imaging unit offers great opportunities to multiplex and/or normalize homogeneous assays. Brightfield plus various fluorescent channels offer a great variety of additional assays such as determining cell confluency e.g. to assess cytotoxicity or check for compound precipitation, or count nuclei after Hoechst staining before lysing your cells for the homogeneous assay.

Simplifying Microtissue-Based Screening Using Automation and the EnSight Multimode Plate Reader
Three-dimensional (3D) cell culture spans the gap between the traditional “petri dish-based” format and whole-animal systems giving unique perspectives on tumorigenesis. Process standardization is essential to enable the generation of robust and reproducible data with more biologically relevant, complex, 3D models. Here, we demonstrate how producing and characterizing 3D spheroid microtissues with the InSphero GravityPLUS™ hanging drop microplate platform can be simplified and standardized by integrating automated liquid handlers with the EnSight™ Multimode plate reader to reduce variability, hands-on time and errors produced by manual pipetting.
Rapid, Reliable Measurement of Cellular Proliferation and Toxic Compound Effects on 3D Spheroid Cultures Grown from Multiple Cancer Cell Lines with ATPlite™ 3D and ATPlite 1step 3D
Cellular viability and toxicity assays are fundamental tools in the drug discovery process – used to evaluate both the potency of compounds, as well as their toxicity profiles for early drug safety assessment. Cells cultured in 3D microtissues exhibit inherent differences in cellular adhesion and require substantially stronger lysis conditions than is sufficient for traditional monolayer or suspension cultures to access the cells deeper inside each spheroid. In order to address this need, we developed ATPlite 3D and ATPlite 1step 3D assay kits specifically for use with 3D cell cultures.
Multiparametric Analysis of Cytotoxicity using Cell Imaging in Combination with Luminescence
Cytotoxicity can be caused by different types of agents, including drugs, pathogens, immune cells and external stress factors such as heat. Particularly during the drug development process, where cytotoxicity remains one of the major causes of drug withdrawal, there is an urgent need for reliable and time-saving assay workflows that gain a more complete picture of the effects of cytotoxicity. This application demonstrates how multiparametric analysis of cytotoxicity, using different technologies, can be easily performed on the EnSight™ Multimode Plate Reader and how brightfield confluency analysis serves as a robust indicator for toxicity in adherent cells and can be easily applied in labeled or label-free cell-based assay.
