Western blot imaging

After purchasing this product several years ago to save money on imaging of western blots, I was recently even more pleasantly surprised. During this time with many company shutdowns, I realized a recent computer replacement had rendered me without the proper software. I was immediately able to reach a representative by phone and received a link to reinstall within 5 minutes. I even had to call again (due to my own error), and was connected to the same person for help. After more than five years, the product and software still work beautifully, and have been used by not only my lab, but others in the department, when their other systems are down. The no-hassle costumer service remains excellent.

Great western blotting imager. Very easy to use!

Western blotting

I run many mini western blots and use the C-DiGit to image the blots. It is very easy to use, very small, and always produces a quality image. The one thing that I would change would be increaseing the imaging surface area so that it can accommodate larger blot sizes (although there is probably another model for that). Overall I am very happy with the ease of use of this product and the results it produces!

I love this machine - small but effective. I made several hundred of western blots and working well, it is easy to use and looks nice on the office table as well :) Also, the customer care is very nice - the people from LiCor are coming time by time and they can also solve some western-problems, so I am very happy with it. The only disadvantage is the software, which do not run on the new versions of Windows - the drivers are no available, so, then it was little problem if the windows is updated.

Great machine, but requires knowledge of its limitations to get the most out of it

Western blots

A lot of the reviews on this site seem like they're written by a marketing company. Anyway, here's a candid view from someone that has regularly used this for two years. For one, the machine is very easy to use. So why did I rate it 3 stars in that category? Well, people who routinely do very long exposures on film are going to wonder why it's not working out for them, and I don't think Licor has been forward enough about the sensitivity, which, in my view, is 40-60% less sensitive than film. We had a lot of people try the machine once and swear to never use it again. An 11 minute exposure on the machine, even when a blot is directly placed on the machine with no plastic wrap, is indisputably not equivalent to an 11 minute exposure on film. That being said, this problem is easily overcome by using stronger substrates (which can also be diluted with water to your likeness!) and/or by improving their western blot technique. It would be wise for Licor to be more forward about this and help users view their fussy blots on the machine from the get-go. They have resources, but they're too minimalistic. Once you know the limitations and tools available, there shouldn't be very many blots you can't get to work on this. I've successfully blotted HIF-1alpha from a whole cell lysate on this machine, which is notoriously difficult to detect. And at the end of the day, you'll get more accurate quantitative data than you would with film.

Antibody Development

We are using the C-DiGit Blot Scanner routinely in our laboratories for the development and characterization of novel antibodies. Setting up the scanner is straightforward and it takes only little time to get started and to produce high-quality western blot images. The software interface is very easy to use. In particular, we like the possibility to choose different "exposure" times. This allows us to screen several antibodies on one single blot in a fraction of time compared to using several ECL films and traditional film development methods. The instant blot digitization on the scanner and the ability to perform quantification further eliminates the need for other equipment and software. For us, the C-DiGit is the perfect solution as it reduces our costs for reagents as well as waste disposal and saves significant space. We therefore highly recommend this scanner for western blot applications.

This is a great little scanner. We find this helps with faster throughput and is certainly cheaper and easier than film. The sensitivity is very good, so that we only occasionally need to go back to film for a particularly fussy antibody.

Western Blot Analysis: Comparison of Film and the C-DiGit® Blot Scanner

Two critical variables affect the accuracy of Western blot analysis: detection chemistry and signal capture method. This study compares the performance of two signal capture methods: film exposure, and the C-DiGit Chemiluminescent Western Blot Scanner. A light-generating LED imaging target was used to eliminate detection chemistry as a source of variability.

Imaging Chemiluminescence by Scanning Using the C-DiGit® Chemiluminescent Western Blot Scanner

The C-DiGit® Chemiluminescent Western Blot Scanner is a digital replacement for film, combining the sensitivity of film with the convenience and flexibility of a CCD imager. To achieve this breakthrough at an affordable price, LI-COR implemented a new imaging approach that was previously considered impossible: line-read scanning of chemiluminescent signals. Film-quality Western images are digitally captured in as little as 6 minutes, and no changes are generally required in detection protocols or reagents. The low-noise sensor array and short optical working distance maximize the efficiency of light collection and enable rapid line-scan imaging. The sensitivity, dynamic range, and affordability of the C-DiGit Blot Scanner make it a true digital replacement for film in Western blotting.

Human Breast Cancer Metastases to the Brain Display Human Breast Cancer Metastases to the Brain Display GABAergic Properties in the Neural Niche

Dispersion of tumors throughout the body is a neoplastic process responsible for the vast majority of deaths from cancer. A rare Investigation of Breast-to-brain metastatic tissue and cells displayed a GABAergic phenotype similar to that of neuronal cells. The GABAA receptor, GABA transporter, GABA transaminase, parvalbumin, and reelin were all highly expressed in breast cancer metastases to the brain. Images were taken on C-DiGit Chemiluminescent Western Blot Scanner.

The C-DiGit Blot Scanner Used in Breast Cancer Metastases Investigations

Dispersion of tumors throughout the body is a neoplastic process responsible for the vast majority of deaths from cancer. Bidirectional interplay of breast cancer cells and native brain cells in metastasis is poorly understood and rarely studied. We had the rare opportunity to investigate uncommonly available specimens of matched fresh breast-to-brain metastases tissue and derived cells from patients undergoing neurosurgical resection. Using the C-DiGit chemiluminescent Western blot scanner (LI-COR Biosciences), among other techniques, we found breast-to-brain metastatic tissue and cells displayed a GABAergic phenotype similar to that of neuronal cells. The GABAA receptor, GABA transporter, GABA transaminase, parvalbumin, and reelin were all highly expressed in breast cancer metastases to the brain. The results suggest that breast cancers exhibit neural characteristics when occupying the brain microenvironment and co-opt GABA as an oncometabolite.
