Top-down Characterization of Monoclonal Antibody on an Orbitrap Fusion Lumos™ Tribrid Mass Spectrometer

8 Mar 2017

This application note describes a method for mass-analysis and top-down characterisation of a monoclonal antibody, conducted on a modified Orbitrap Lumos mass spectrophotometer. LC-MS analysis was performed on the intact monoclonal antibody, the IdeS sub-units and the reduced heavy chain product. ETD HD, UVPD and CID top-down fragmentation modes were applied to obtain the optimal sequence coverage.

Thermo Scientific™ MAbPac™ RP LC Columns

Thermo Fisher Scientific

Use the new Thermo Scientific™ MAbPac™ RP columns for high-performance reversed-phase (RP) HPLC and LC/MS characterization of monoclonal antibodies (mAbs), fragments, variants, antibody drug conjugates (ADCs), PEGylated proteins, and bispecific proteins. These columns have excellent resolution, long column lifetime, and low carry-over. The surface chemistry, particle size, and pore size, have been optimized to for high resolution separations. Compatible with organic solvent used in HPLC and MS under RP conditions Resin is based on a hydrophobic supermacroporous 4µm polymer particles Stable at extreme pH (0 to 14) and high temperature (up to 110 °C) Efficient with low carry over 1,500 Å large pore size polymeric resin; Efficient separation of protein molecules with very low carry over Designed for reversed-phase biomolecule LC and LC/MS separations Superior resolution power for monoclonal antibodies and related substances High efficiency with low carry-over Excellent MS compatibility Wide operating pH range: 0 to 14 High temperature stability: up to 110 °C High throughput
