Successful Use of Thermo Scientific miRIDIAN microRNA Mimic and Inhibitor Libraries Identify microRNAs Involved in Human Mesenchymal Stem Cell Osteogenesis

11 Nov 2013

MicroRNAs are endogenous non-coding RNAs that modulate gene expression through the RNA interference pathway. Human mesenchymal stem cells (hMSC) are capable of differentiating into multiple lineages. Unfortunately, while these cells can be easily isolated and expanded, little is known about the contributions that microRNAs make to MSC differentiation. The following application note demonstrates the validity of how screening with miRNA mimic and inhibitor libraries can be successfully used to identify microRNAs that play a role in cell fate determination, specifically, hMSC osteogenic differentiation.

DharmaFECT 1 Transfection Reagent

Thermo Fisher Scientific

Efficient siRNA or microRNA transfection. To attain efficient and reliable siRNA or microRNA transfection, we offer DharmaFECT transfection reagents. Thermo Scientific DharmaFECT 1 is our most all-purpose transfection reagent formulation; demonstrating efficient, low-toxicity delivery to over 35 cell types. DharmaFECT 1 achieves effective transfection of siRNA and microRNA reagents at low concentrations and with low cytotoxicity, especially when compared to transfection reagents designed for use with plasmid DNA.While DharmaFECT 1 is the most all-purpose transfection reagent (demonstrating efficient, low-toxicity delivery to over 80% of validated cell types), DharmaFECT 2, 3, and 4 are available to support a wider range of cell types. Distinct formulations permit more thorough optimization of transfection for novel cell types, high-value experiments, and RNAi screening.DharmaFECT 1 Transfection Reagent Features: Optimized for small RNA (siRNA or microRNA) delivery at low concentrations Low toxicity – Maintain cell viability with lipids specially formulated to deliver siRNA Highly efficient delivery – Achieve robust siRNA uptake for reliable gene silencing
