Gentle Sorting of PBMC Populations

8 Feb 2019

Successful cell sorting via flow cytometry highly depends on the collection of viable cells. It is well established that high pressure, decompression, and shear forces negatively affect cell viability and functionality during conventional droplet sorting. Therefore, to mitigate the risk of cell damage, instrument settings, including nozzle size and pressure, have to be carefully chosen by experienced operators, who are capable of adjusting the settings accordingly. Sterility is another concern when working with traditional cell sorters that often operate in an uncontrolled, open-air surrounding. Samples are prone to contamination from both internal sources (sheath fluid, sampling fluidics, etc.) and external sources (airborne contaminants).

MACSQuant® Tyto® Cell Sorter

Miltenyi Biotec B.V. & Co. KG

The MACSQuant ® Tyto ® is a next generation, benchtop cell sorter equipped with 3 lasers, which allows for high speed, 10-parameter cell sorting. A unique feature of the instrument is the fact that the actual sorting process takes place exclusively within a single-use, disposable and fully closed system: the MACSQuant Tyto Cartridge. The closed cartridge provides full operator safety and eliminates the risk of sample contamination and carry-over.

Flow Cytometry / Cell CountingFlow cytometers are used to count, sort and examine multiple characteristics of cells. Other cell analysis equipment includes image cytometers, cell counters, fluorescence-activated cell sorters (FACS), magnetic-activated cell sorters (MACS), and a range of flow cytometry assay kits. Flow cytometers can reveal information on cell viability, cell proliferation, apoptosis and cell cycle progression, as well as identify cell populations and intracellular or cell-surface molecules. Additionally, some flow cytometers, known as FACS, have an additional sorting function after analysis. Cell counters and image cytometers count live and dead cell populations and can also conduct cell proliferation assays.
