Biochemical Characterization of an Enzyme by Calculation of Km and Vmax according to the Model of Michaelis and Menten

This application note explains the use of TECAN Magellan software together with a TECAN SUNRISE microplate reader for spectrophotometric measurements of an enzymatic assay of alkaline phosphatase in glycine buffer. From the kinetic raw data, the kinetic parameters, maximum reaction velocity Vmax and the Michaelis constant Km, are calculated.
