This application note describes the accurate PCR amplification, of fragments from the β-globin, K-ras 12 and p53 genes in DNA extracted from sections of 10-year-old paraffin-embedded colon cancer tissues, using TaKaRa Ex Taq™.

This Takara Bio SYBR Green Master Mix is recommended for real-time PCR (qPCR) when high specificity is critical, because this reagent effectively suppresses amplification of primer-dimers and off-target products. Use this product when primer design has been optimized, but an increase in specificity of the qPCR assay is still needed. SYBR Premix Ex Taq II (Tli RNase H Plus) includes SYBR Green I, a reagent designed for intercalator-based qPCR. Additionally, SYBR Premix Ex Taq II (Tli RNase H Plus) is supplied at a 2X concentration, enabling easy reaction assembly. The premix also contains heat-resistant Tli RNase H Plus, which effectively degrades residual mRNA found in the input cDNA, thereby minimizing qPCR inhibition as a result of residual mRNA presence.Takara SYBR Green Master Mix products contain TaKaRa Ex Taq HS DNA Polymerase, a hot start PCR enzyme that includes an anti-Taq antibody. The use of an anti-Taq antibody allows highly reproducible and reliable qPCR.
