Point-of-care monitoring for drugs of abuse

Therapeutic Drugs of Abuse testing for critical point-of-care treatment, such as in drug treatment facilities or pain clinics, has traditionally been arduous, expensive and time consuming, requiring a third-party laboratory for analysis. Due to these factors, on-site liquid chromatography (LC) analysis at the local clinic is a requisite for timely evaluation and effective treatment. A point-of-care LC system must be easy to use, self-contained, safe for the user, hand portable, and compact to be effective. In this application note, Axcend shows how the innovative Axcend Focus LC® (AFLC) is well suited for point-of-care Drugs of Abuse testing.

12-hour dissolution of an Extra Strength Excedrin brand analgesic in 200ml of media with the Axcend Focus LC

The Axcend Focus LC® High-Performance Liquid Chromatography system (HPLC) delivers repeatable analyses of Active Pharmaceutical Ingredient (API) dissolution. In this application note, an Extra Strength Excedrin® analgesic tablet was dissolved, without stirring, in a beaker containing 200 mL of media [97% water; 3% acetonitrile (ACN); and <0.1% trifluroacetic acid (TFA)]. With samples acquired approximately every 12 minutes, the three APIs (acetaminophen, aspirin, and caffeine) were well separated at 4.5 min/run with an average retention time of 2.75 min, +/- 0.05 min RSD (relative standard deviation). Additionally, only 513 µL (0.51 mL) of organic and aqueous solvents were consumed during the 12-hour experiment, resulting in dramatically lower costs for solvents and waste disposal.

Two methods for quick analysis of polycyclic aromatic hydrocarbons (PAHS) using the Axcend Focus LC

In this application note, the Axcend Focus LC® was used to separate polycyclic aromatic hydrocarbons (PAHs) in ~12 and ~15 minutes, respectively, with separate methods on short (10 cm x 150 µm i.d.) capillary columns packed with 1.7 µm monomeric and 3.0 diameter polymeric C18 particles. As a lightweight, hand-portable, AC- or DC- powered high-performance liquid chromatography system that utilizes capillary columns and on-column UV-absorption detection, the Axcend Focus LC is uniquely suited for both in-the-field or laboratory-based HPLC detection, separation and analysis of a variety of target chemicals, including products and by-products of fossil fuels, pollutants, and agrichemicals, to name a few.

Intact protein analysis by RPLC using hand-portable HPLC

In this application note, Axcend demonstrates the use of hand-portable capillary HPLC by the Axcend Focus LC for intact protein analysis. Specifically, the separation of five test proteins in the range of 5.8 to 45 kDa is shown using reversed phase (RP) conditions. Other protein samples can be analyzed using the Axcend Focus LC merely by adapting established conventional high performance LC methods to the capillary column format.
